Objective :To research the role of Mfn-2 expression and PI3K-Akt signaling pathway in reduction of myocardial ischemia-reperfusion injury by sevoflurane postconditioning in rats.Methods :Thirty male Sprague-Dawley rats were randomly divided into 5 groups(n=6):group SH(Sham operation group),group MIRI(ischemia-reperfusion group),group SP(sevoflurane postconditioning group),group SP+W(W is short for Wortmannin,a specific PI3 K inhibitor),and group W.After anesthesia and fix the rats,except group SH,the rest of the four groups would be ligation tight the left anterior descending branches for 30 minutes,then recover blood flow for 120 minutes.Group SP and Group SP+W should inhale 2.3% sevoflurane for 15 minute s,when it is 10 minutes before the blood flow recovered.Group SP+W should be given a 15ug/kg diluented Wortmannin by vein injection 15 minutes before the blood flow recovered.Group W is given the diluented Wortmannin only 15 minutes before the blood flow recovered.Recorded the entire ECG.Then measure these indicators: cTn-I with ELISA.Total Akt,P-Akt and Mfn-2 expression with Western Blot,calculate the ratio of the P-Akt and total Akt,also contrast the morphology changeof myocardial cell underopticalmicroscopy.ResultsThere is no statistically significant difference among all the groups of total Akt(P>0.05).Compared with group SH,the other groups are increased statistically significant in aspect of cTn-I(P<0.05).Group SP is less than group MIRI,group SP+W and group W in aspect of cTn-I,there is a statistically significant difference(P<0.05).Among group MIRI,group SP+W and group W in aspect of cTn-I,ther eis no statistically significant difference(P>0.05).Compared with group SH,the other groups are increased statistically significant in aspect of P-Akt(P<0.05).Group SP is more than group MIRI,group SP+W and group W in aspect of P-Akt,there is a statistically significant difference(P<0.05).Among group MIRI,group SP+W and group W in aspect of P-Akt,there is no statistically significant difference(P>0.05).Compared with group SH,the other groups are increased statistically significant in aspect of Mfn-2(P<0.05).Compared with group SP,group MIRI and group W are increased statistically significant in aspect of Mfn-2(P<0.05).There is no statistically significant difference between group SP and group SP+W(P>0.05).There is no statistically significant difference between group MIRI and group W neither(P>0.05).It is showed that there are no signs of damage in group SH,while group SP shows a significant damage lighter than group MIRI,group SP+W and group W by the optical microscope.Conclusion :1.Sevoflurane postconditioning can alleviate the myocardial ischemia-reperfusion injury in rats,probably via with the reduction of Mfn-2 expression and activate the PI3K-Akt pathway.2.In this experiment of alleviate MIRI,the reduction of Mfn-2 expression is not at the downstream of the active PI3K-Akt pathway.
