Objective:1.To investigate the effects of different concentrations of angiotensin ? on autophagy related protein expression of cardiomyocyte,Akt and ERK signal pathway.2.To investigate the inhibition of autophagy effect on angiotensin ? induced cardiomyocyte hypertrophy signal pathway.Menthods:1.Estabish H9C2 cardiomyocytes culture models,CO2 content of 5%,37? the incubator and cultured.2.Groups:(1)normal control group(2)angiotensin ? intervention(3)angiotensin ? intervention and antophagy inhibitors(3MA).3.Methods:By Western Blot to detect protein expression in cardiomyocytes,using Image Lab5.0 and Image J11.0 to analysis the grey value of strips.Results:Compared with the control group(0 nmoL/L),Ang?(100nmoL/L)intervene cardiomyocytes 1hour,the phosphorylation of ERK(p-ERK)protein expression increased significantly(P<0.001),the phosphorylation of Akt(p-AKt)increased no significantly(P>0.05).ERK?AKt protein expression was not significant change(P>0.05).Autophagy inhibitor 3MA(5mmoL/L)could reduce p-Akt(P<0.001)protein expression by Ang? and p-ERK protein expression increased(P<0.05).Further research result show that compared with the control group,Ang ? induced autophagy related Beclin1 protein expression increased,3MA inhibit the Beclin1 expression induced by Ang?(P =0.005),there was statistical significance;LC3 expression decreased slightly(P>0.05),there was no statistical significance.Conclusion:Ang? induced cardiomyocyte autophagy protein increased,Akt and ERK activity.Autophagy inhibitor 3MA,inhibit the AKt and ERK induced by Ang?.The study suggests inhibit cardiomyocyte autophagy could reduce cardiomyocyte hypertrophy signal pathway,thus alleviate cardiomyocyte hypertrophy,inhibit autophagy is beneficial for the treatment of cardiomyocyte hypertrophy.
