Objective:A multiplex amplification system for individual identification in Chinese populations was established by using insert-deletion(InDel)polymorphism genetic markers,and the system was applied to forensic biological evidence examination.Methods:The multiplex amplification system was constructed by the following steps including,the selection of the InDel loci,the screening of the fluorescent dyes,the design adjustment of the primers,the optimization of the amplification system and the reaction conditions.The amplification system was validated by stability,sensitivity,species specificity,tissue identity and forensic sample test.Finally,a population genetics survey was conducted in the following populations like Han,Miao,Kazakh,Yao,and Dai,in order to validate the effectiveness of the present system in individual identification.Results:On the basis of a large number of experimental studies,a multiplex system was established 30 InDel loci and a gender identification locus,a total of 31 loci in the system.The system has good stability and sensitivity to different samples,and can detect and type degraded materials accurately and efficiently.In the population genetic survey,the cumulative discrimination power of the Han nationality,the Miao nationality,the Kazak nationality,the Yao nationality and the Dai nationality were 0.999999999957,0.99999999875,0.999999999990,0.99999999966,0.99999999974 respectively,which were comparable to the Qiagen commercialization kit.The amplification reagents,buffer,are self-developed which reducing the detection costs.The system can be applied to the Chinese population individual identification and can be used as an effective complementary part of STR typing method.Conclusion:The system can be used for forensic DNA individual identification in Chinese population.The system can be used to test different kinds of forensic biological evidence including degraded DNA.
