Objective To detect the expression of HAS2 gene in human cumulus cells from patients receiving intracytoplasmic sperm injection-embryo transfer(ICSI-ET)treatment in our center and investigate the correlation between the expression of HAS2 gene and oocyte maturation and embryo development potency.Methods During August2014 and December 2015,there were 45 infertile patients receiving ICSI-ET treatment in our center due to male factor infertility.Clinically,a same long-term long program was used to conduct superovulation.Oocytes were obtained by ultrasound-guided puncturing.Among them,150 of the oocytes were retrospectively analyzed.The surrounding cumulus cells of the 150 oocytes were stripped and cumulus cells were collected individually.The maturity,fertilization,embryo score and blastocyst formation situation of the corresponding oocytes were observed.According to the maturity of the oocytes,the oocytes were divided into mature oocyte group(n = 100)and immature oocyte group(n = 50).According to the embryo morphology scoring standard,the normally fertilized D3 embryos were divided into high quality embryo group(n = 28)and non-high quality embryo group(n = 72).After transplant,the remaining embryos were subjected to cryopreservation and blastocyst culture.According to the situation of D5/D6 blastocyst culture,it were divided into the successful culture group(n=26)and the failure group(n=22)Real-time fluorescent RT-PCR was used to detect the expression level of HAS2 gene in cumulus cells.The differences of HAS2 gene expression levels among the three groups were compared.Results In the mature oocyte group,the expression level of HAS2 was significantly higher than that in the immature oocyte group(2.4707 ± 0.89738 vs.1.8555 ± 0.87439,P < 0.05);In the D3 high-quality embryo group,the expression level of HAS2 in cumulus cells was statistically significant higher than that in the non-high-quality embryo group(5.1179 ± 1.83082 vs.2.8964 ± 0.60215,P < 0.001).Further blastocyst culture found that HAS2 gene in D5/D6 blastocyst successfully cultured cumulus cells was higher than that in the blastocyst unsuccessfully cultured cells(4.5654 ± 1.3997 vs.1.9885 ± 0.62758,P < 0.05).The results were analyzed by SPSS 17.0.Software.experimental data were performed by t tests and variance analysis.P<0.05 was considered statistically significant.Conclusions In the mature oocytes,D3 high-quality embryos and D5/D6 blastocyst,the HAS2 expression levels were relatively higher,suggesting that the expression level of HAS2 in cumulus cells may reflect the maturity of the oocytes and embryo quality.By detecting the HAS2 expression levels in the oocytes,the ICSI embryo development potency may be predicted.
