Objective To explore the relationship between spinal cord injury(SCI)and myocardial energy metabolism by the gene expression analysis of mitochondrial oxidative phosphorylation which will clarify the possible mechanism of myocardial injury after SCI.Method Thirty clean male Sprague-Dawley rats weighing 250-300 g were used.And they were randomly divided into five groups:the sham-operation control group(TC group)and the experimental group(TS group): high spinal cord injury after myocardial extraction time for 3 h group(TS1 group),12 h group(n = 6),including sham operation group(TC group)(TS2 group),24 h group(TS3 group),48 h group(TS4 group).The rats were treated with modified Allens beat method,and the myocardium tissue of the same apex was taken at each time point for the further reach.The ultrastructure of myocardial cells was observed by transmission electron microscopy.At the same time,84 genes involved in the mitochondrial oxidative phosphorylation in myocardial energy metabolism were analyzed by PCR assay gene expression analysis.Result(1)The results of transmission electron microscopy showed that in the TC group the nuclei of myocardium were intact,the myofilament was intact and the filaments were intact.The cell membrane was complete and the mitochondrial ridge was clear.However in the TS group,there were vacuoles,cell necrosis and other changes in the myocardium.The cell membrane was partially damaged,the chromatin hatching and the cell vacuolar structure appeared.The mitochondria appeared obviously Shift,concentrate.(2)PCR assay results: In the 84 genes detected,compared with the TC group,TS group showed a certain degree of oxidative phosphorylation related gene expressionchanges,there are differences in the expression of a total of 18 genes.Among them,there were significant changes in the expression of genes in the TS4 group.Atp4 a,Atp6ap1,Cox17,Cox4i2,Cyc1,Ndufa8,Ndusb6,Ndufs1,Sdhd,Sic25a10,Ucp2,Ucp3 and Uqcrfs1 show an upgrade expression(ie,Fold value> 2)in total of 13.A total of five genes show an downgrade expression whiche include: Atp6v0a2,Atp6v1e2,Bcs11,Cox15 and Lhpp.Compared to the TC group,in the TS group Atp6v1e2,Lhpp and Sic25a10 were up-regulated at 3 h,12 h,24 h and 48 h after injury.The expression of Atp6v1e2 in TS group was up-regulated particularly at 24 h,and the expression of Lhpp was up-regulated at 24 h.Sic25a10 was down-regulated most significantly at 3 h,and rebounded at 12 h,but still lower than the TC group in each time point.In addition to Cox15 up-regulated in 24 h,48 h group and Ucp2 upregulation at 3 h,24 h expression,the rest of the gene expression were all down-regulated at 48 h.Conclusion 1.The myocardium ultrasonography injury was observed in rats with high spinal cord injury.2.Myocardial injury may be related to mitochondrial oxidative phosphorylation of electron transport chain damage,which may be related to ATP synthase(Atp4a,Atp6ap1),complex?(Ndufa8?Ndusb6?Ndufs1)??(Sdhd)??(Cyc1)??(Cox17?Cox4i2)and other related genes.3.The changes in oxidative phosphorylation gene expression may not be the major cause of the damegement of myocardium in rats with high-level SCI.
