A Study On The Correlation Between Rs505151/rs2479409 Polymorphisms Of PCSK9 Gene And Coronary Heart Disease In Fujian Han Population

Objective:Examined the distribution of rs505151 and rs247940 polymorphisms of the pre-protein converting enzyme subtilisin-9(PCSK9)gene in the Han population of Fujian,China,and analyzed the correlation between the polymorphisms and CHD.Methods:Blood samples selected from 184 cases of CHD diagnosed by coronary angiography and 60 cases of Non-coronary heart disease were studied.There was no significant difference in age and sex between the two groups.The gene polymorphism of rs505151 and rs2479409 in PCSK9 gene were detected by PCR—RFLP.Collected the body mass index(BMI),blood glucose,blood pressure,smoking history,hypertension history,diabetes history etc.The levels of TC,TG,LDL-C,HDL-C,Apo A,Apo B,Lp(a)and FBG were measured by enzymatic methods.Results:1.Smoking rate,prevalence of hypertension and diabetes,as were as the level of TC,HDL-C,LDL-C and ApoB were statistically significant between CHD group and control group(P <0.05).The level of HDL-C in CHD was lower than the other group.2.The rs505151 locus of PCSK9 gene:(1)The genotypes of AA,AG and GG were detected.The genotype frequency of AA,AG and GG in CHD group were 80.98%,16.30% and 2.72%,the allele frequency of A,G were 89.13% and 10.87%.The genotype frequency of AA,AG and GG in control group were 96.7%,3.33% and 0.00%,the allele frequency of A,G were 98.33 % and 1.67%.There were significant differences of genotypeand allele frequencies between the two groups(P <0.05).(2)There were significant differences in TC and LDL-C levels between the three genotype in the CHD group(P <0.05).And the levels of TC and LDL-C were higher in patients with G allele(GG > AG > AA).(3)The Gensini scores of AA,AG and GG were 45.06 ±27.50,57.58 ± 25.44,83.80 ± 24.00.There were significant differences between the three genotype and the Gensini score.Gensini score were higher in G allele carriers(GG >AG > AA)..There was no significant difference in gene distribution among different groups of coronary artery lesions.3.The rs2479409 locus of PCSK9 gene:(1)The genotypes of AA,AG and GG were detected.The genotype frequency of AA,AG and GG in CHD group were 11.96%,43.48% and 44.56%,the allele frequency of A,G were 33.70% and 66.30%.The frequencies of AA,AG and GG were 10.00%,41.67% and 48.33% in control group,the allele frequency of A,G were 30.83% and 69.17%.There was no significant difference of genotype and allele frequencies between the two groups(P> 0.05).(2)There was a significant difference in TG level between AA and GG in CHD group,and a significant difference in LDL-C level between AG and GG genotypes.(3)The Gensini scores of AA,AG and GG genotypes in CHD group were 40.55 ± 25.95,51.03 ± 28.04 and 47.99 ±27.49,there were no significant differences between the three genotypes(P> 0.05).(4)There was no significant difference in gene distribution among different groups of coronary artery lesions.Conclusions :1.The frequency of G allele in rs505151 locus of PCSK9 gene in Fujian Han population was higher in coronary heart disease population,and the level of TC and LDL-C as were as the degree of coronary lesion were higher in patients with G allele.2.There was no difference in the rs2479409 polymorphism of the PCK9 gene between the coronary heart disease population and non-coronary heart disease population.The rs2479409 gene was related to the higher level of TG,LDL-C,but not related to the degree of coronary artery disease.
