Objective Using immunohistochemical method,to explore BMP-2 and VEGF expression in HCC,combined with the patient clinical pathology data,to analysis the expression of them in HCC.at the same time CD34 markered tumor microvascular in the tumor tissue, combined with the expression of BMP-2,VEGF and CD34 in HCC and the relationship of the clinical pathological data,to explore the relationship BMP-2 ?VEGF and tumor angiogenesis,BMP-2 plays a key role in angiogenesis in HCC.It provides a new treatment approach.Method40 HCC specimens and paired non-tumor tissues were collected from 40 HCC patients undergoing surgery operation during June 2015 to June 2016 in Tianjin third central hospital. All patients were diagnosed as HCC by HE staining. All patients underwent operation for the first time.No one have underwent adjuvant therapy such as radiotherapy and chemotherapy before the operation. The expressions of BMP-2and VEGF in HCC were analyzed immunohistochemical streptavidin-peroxidase method to assess the relationship between BMP-2 and VEGF with tumor angiogenesis.Results1.The expression of BMP-2 and VEGF were located in cytoplasm of HCC tissue. The positive rate of BMP-2 and VEGF in HCC tissues(75%and 80%,respectively)were higher than that in paired non-tumor tissues(40% and 42.5%,respectively),and the difference were statistically significant(Chi-square value:10.03 and 11.85,respectively,P<0.05).2.The expression of BMP-2 and VEGF weren't related to age, sex,AFP,HBV,cirrhosis and the size of the tumor. However, they were related to the integrity of HCC tissue, the number of nodules, portal vein tumor thrombus,TNM and degree of differentiation.3.Based on the Spearman, the expression of BMP-2 was positively correlated with VEGF(r=7.316,P=0.0068).BMP-2 and VEGF were positively correlated with tumor angiogenesis. The expression of CD34 in HCC tissues was higher than that in paired non-tumor tissues(55% and 15%,respectively.P <0.05).Conclusions1.The expression of BMP-2 and VEGF in HCC tissues is higher than that in paired non-tumor tissues. And the expression of BMP-2 is positively correlated with VEGF.2.The expression of BMP-2 and VEGF aren't related to age, sex,AFP,HBV,cirrhosis and the size of the tumor. However, they are related to the integrity of HCC tissue, the number of nodules, portal vein tumor thrombus,TNM and degrees of differentiation. The higher the BMP-2 is the more prone to invasion and metastasis of HCC.The expression of BMP-2 is positively correlated with tumor angiogenesis, which provides a good method of anti-angiogenic therapy.
