Objectives Fluorescent probe SLeX(sialylated Lewis oligosaccharide-X antigen)was used to detect serum samples,and a new method for the diagnosis of prostate cancer was established to improve the early diagnosis rate of prostate cancer.To explore the value of fluorescent probe SLeX in the diagnosis of prostate cancer and its significance in clinical TNM stage,pathological grade and Gleason score.Methods A total of 76 patients with urological hospitalization were enrolled in the hospital from January 2015 to December 2016.Among them,38 patients(BPH group)and 38 patients(PCa group)of benign prostatic hyperplasia were treated with prostate biopsy or/And postoperative pathological diagnosis.(T1,T2)in 16 cases and non-limited group(T3,T4)in 22 cases;by Gleason score(2 to 4 subgroups)were divided into two groups according to the American Cancer Society(AJCC)TNM staging in 2002 High differentiation of cancer;5 to 7 is divided into differentiated carcinoma;8 to 10 divided into poorly differentiated carcinoma)is divided into well differentiated carcinoma,moderate differentiation of cancer,poorly differentiated carcinoma were 12 cases,10 cases,16 cases.The expression of PSA-SLeX(prostate-specific sialylated Lewis oligosaccharide-X antigen)in serum was detected by ELISA(ELISA)with the fluorescent probe SLeX as the detection antibody.Detection of serum TPSA expression by double-antibody sandwich method and comparative analysis of the value of the two detection methods in the diagnosis of prostate cancer and its significance in clinical TNM stage,pathological grade and Gleason score Feasibility and significance..Result1.PSA-SLeX was expressed in prostate cancer and benign prostatic hyperplasia,but was highly expressed in prostate cancer,and there was a significant difference between the two groups(P = 0.000).2.Using the ROC curve,the area under the curve of PSA-SLeX and TPSA was 0.851 and 0.815,respectively.Fluorescence probe SLeX was equivalent to the diagnostic value in TPSA prostate cancer(P>0.05).Serum PSA-SLeX and TPSA in the prostate cancer test to determine the optimal cut-off value(ie cut off value)were 13.15ng/ml and 9.68ng/ml.3.The concentration of PSA-SLeX in TNM staging of prostate cancer was higher than that of TPSA(P = 0.001).4.In the Gleason score of prostate cancer,the concentration of PSA-SLeX in serum of middle and poorly differentiated prostate cancer group was higher than that of TPSA,the difference was statistically significant(P<0.05).Conclusions:Fluorescent probe SLeX was used in the screening of clinical prostate cancer,and the positive rate of poorly differentiated detection was superior to that of TPSA in the late stage of TNM staging of prostate cancer and Gleason score.
