Objective:1.To study the effect of different oils on blood lipids,oxidative stress and endothelia function among mice.To explore the relationship between oil supplements and cardiovascular health in order to further study oil product development.2.To analyze the difference effects between mixed oil(soybean oil:palm oil 0.9:1)and soybean oil.3.In order to study the effect of different oils on the intestinal flora and guide on the selection of oil in everyday life,the experiment was taken on the basis of the first chapter.Methods:1.96 C57BL/6 mice were divided into 8 groups.The average weight of C57BL/6 mice was from 16g to 18g.Of 7 groups were fed with different high-fat diet per day,including lard,ester exchange oil,soybean oil,24 degrees palm oil,33 degrees palm oil,soybean oil,(palm oil:soybean oil 0.9:1)mixed oil and olive oil,1 group was set as control group fed with chow diet.After 12-week,blood samples were collected and then the mice were euthanized.The adipose tissue is preserved in formaldehyde solution.The left tissues were pretreated to detect indicators of serum lipids,oxidative stress and endothelial cell function.2.Two different postprandial lipid metabolism experiments were taken on healthy person in Southeast University.There was one month interval between two experiments.The first postprandial lipid metabolism experiment was mixed oil(soybean oil:palm oil 0.9:1),and the second postprandial lipid metabolism experiment was soybean oil.After a 10-12 hours overnight fast,fasting venous blood samples were gathered by an indwelling needle fitted in the subjects'cubital veins.Following this,a liquid high-fat meal was given to be ingested in 5 minutes.The blood samples were collected after 2,4,6 hours.Serum lipids,oxidative stress and endothelial cell function indicators were measured.3.After the first chapter,the residual waste was taken from the colon.DNA was exacted from the fresh fecal samples.Gut microbiota analysis were investigated based o n the sequencing results on V4 region of 16S rDNA.At last,analyze the OTUs.Results:1.Long-term high fat diet intake caused a rise in serum TC and TG levels.Serum TC level was observed with significant difference between experimental groups and the control group(P<0.05),and the serum TC level of olive oil group was lower than the 33 degrees palm oil and ester exchange oil group(P<0.05).The serum TG level in experiment groups was higher than the control group except the olive oil group(P<0.05).The serum TG level of olive oil was lower than the lard group(P<0.05).The serum MDA level of the experimental group was higher than control group(P<0.05).The serum MDA level of Olive oil and soybean oil group were lower than the lard group(P<0.05).The serum endothelin level of the experimental groups was higher than the control group except the soybean oil group(P<0.05).The serum endothelin level of the soybean oil group was lower than the lard group and 33 de~grees palm oil group(P<0.05).There was no significant statistic difference between 8 groups on the indicators of IL-6,GSH-Px,and TNF-? and so on.2.On the change trend of postprandial serum TG level,there was a significant statistic difference between two groups(P<0.001).The serum TG level of soybean oil group had a rising trend over time.In the mixed oil group,there was a rising trend at 2 hours.Then the level fell at 4 hours,and reached the level of the soybean oil group at 6 hours.There was no significant statistic difference between two groups on the indicators of TC,HDL-C,MDA,SOD,NO,ET-1.3.In Actinobacteria,there was a significant statistic difference between control group and the mixed oil group(q value<0.05).In verrucomicrobia,the control group was lower than lard group and 33 degrees palm oil group(q value<0.01).Conclusion:1.Compared with other oils,olive oil can reduce the blood lipids.Compared with the lard,soybean oil and olive oil can not increase the oxidation level of body,lard are more likely to lead to the oxidative stress level of body.The lard and the 33 degrees palm oil can increase the level of endothelin,and the soybean oil,abundant with polyunsaturated fatty acids,has inhibitory effects on endothelin release.2.There is no difference between soybean oil and mixed oil(soybean oil:palm oil 0.9:1)on the indicators of oxidative stress and endothelia function.There is application prospect on the mix of 24 degrees of palm oil and soybean oil.3.C18:0 can be key factors in oil to cause verrucomicrobia abnormal,which indicates long-term high saturated fat diet may affect intestinal flora.Soybean oil,olive oil,24 degrees palm oil do not change intestinal abnormalities.
