Long-noncoding RNA(1ncRNA)can play biological function through a variety of mechanisms,including gene stenosis and chromatin remodeling,regulating cell cycle and regulatory gene splicing,and regulation of mRNA degradation and mRNA translation.These mechanisms of the effect are at different levels of gene expression regulation.A preliminary study of the laboratory found a new long-chain non-coding RNA gene,we temporarily named Z38.It has been confirmed that LncRNA Z38 gene is highly expressed in tumor tissue and low in adjacent tissues.The aim of this study was to investigate the effect of LncRNA Z38 gene expression on the function and mechanism of human nasopharyngeal carcinoma cell line HNE1 and 5-8F,mainly at the cellular and molecular levels.Specifically carried out the following work:1.Transfection of shRNA-Z38 into nasopharyngeal carcinoma cells HNE1 and 5-8F,through screening of puromycin to obtain stable interference of nasopharyngeal carcinoma cell lines.2.Real-time fluorescent quantitative PCR was used to detect the expression of LncRNA Z38 in nasopharyngeal carcinoma cells HNE1 and 5-8F.3.The proliferation and cell migration ability of nasopharyngeal carcinoma cells treated with LncRNA Z38 were detected by Transwell assay using MTT and plate clone colony formation.4.Western blot was used to detect the expression levels of p53 and p21 in nasopharyngeal carcinoma HNE1 and 5-8F after LncRNA Z38 expression.The results showed that lentiviral vector-mediated RNAi technology significantly reduced the expression of LncRNA Z38 gene and disrupted the expression of LncRNA Z38 gene in nasopharyngeal carcinoma cells after interfering with nasopharyngeal carcinoma cells.The expression of LncRNAZ38 was confirmed at the cellular level,and the proliferation ability and colony forming ability of nasopharyngeal carcinoma cells were decreased and the number of cells passing through the Transwell chamber was decreased in a series of tumor cell functional determination.Western blotting analysis showed that the expression of p53 and p21 in nasopharyngeal carcinoma cells was up-regulated after LncRNAZ38 was interfered.This result is consistent with our hypothesis that the Z38 gene is one of the cancer-related genes.
