Objective:To investigate the expression of mismatch repair protein(MMR)in endometrial carcinoma,to investigate the relationship between the occurrence and development of endometrial carcinoma,and to explore the significance of MMR in the prevention of cancer patients with Lynch syndrome and the family members.Method:The Third Hospital Affiliated to Soochow University from 2011 02 to 2016 09 months in 100 cases of endometrial cancer endometrial tissue specimens,the expression of MMR protein(including MLH1,MLH2,PMS2,MLH6)was detected by immunohistochemical method,and compare it with age,pathological type,differentiation grade,myometrial invasion,lymph node metastasis and clinical stage whether the relationship between.Results:In 100 specimens of endometrial carcinoma,the general expression of MMR protein deletion rate was 22%;MLH1,MLH2,PMS2,MLH6 expression rates were: 13%,5%,14%,7%,MLH1 and PMS2 co expression rate: 13%,MLH2 and MLH6 and expression rate of 4%.In all age groups,30-40 years old group,41-50 years old group,above 50 years old group,the expression of MMR protein deletion rate was 20%(2/10),respectively,with a total of 43.33%(13/30),11.7%(7/60),the difference was statistically significant(P=0.003),the deletion of MMR protein expression was significantly higher in the group under the age of 50 years old than the group above the age of 50;The expression of MMR protein was significantly higher in endometrial adenocarcinoma,and the deletion rate was 22.7%(22/97),which was significantly higher than that of the special endometrial carcinoma group;In the pathological differentiation grade,G1 grade,G2 grade,G3 grade,the deletion rate of MMR protein expression were 10.2%(5/49),23.3%(7/30),47.6%(10/21),the deletion rate of MMR protein in advanced group(G2-G3)was significantly higher than that in low level group(P=0.005);In different degree ofmyometrial invasion,the deletion rate of MMR protein expression was 43.8%(7/16)in the myometrial invasion group(>1/2),was significantly higher than that of <1/2 myometrial invasion group 17.9%(15/84)(P=0.022);In the presence oflymph node metastasis,the deletion rate of MMR protein expression was 71.4%(5/7)in the metastatic group,which was significantly higher than that in the non metastasis group 18.4%(17/93),the difference was statistically significant(P=0.001);In different clinical stages,the deletion rate of MMR protein expression in FIGO I,II,III was 16.9%(13/77),41.7%(4/11),(5/12),there was no significant difference between the three groups(P=0.074),there was no significant relationship between the expression of MMR protein and clinical stage.Conclusion:The expression of MMR protein is closely related to the occurrence and development of endometrial carcinoma.In the age of ?50 years,the high level of differentiation,>1/2 of myometrial invasion of endometrial adenocarcinoma and lymph node metastasis,loss of MMR protein expression rate was significantly increased,butthere was no significant correlation with clinical stage,its detection can be used in clinical screening of endometrial cancer in patients with Lynch syndrome and their family members tumor early prevention.
