| The aim of study:To observe the effects of long-term exercise,trehalose and combination of exercise and trehalose on cognitive function and cortex autophagy signaling in aged mice.To explore the relationship between autophagy activation and exercise and trehalose in improving cognitive function of aging brain.Through scientific fitness to achieve healthy aging and successful brain aging to provide theoretical and experimental basis for the elderly.Methods:40 C57BL/6 male mice aged 15 months were divided into aged control(OC)group,aging trehalose(T)group,aging exercise(E)group,aging exercise +trehalose(E+T)group,and another 10 C57BL/6 mice aged 3 months as young control(YC)group.E group,E+T group of mice for a period of 12 weeks swimming exercise training.After 48 h of the last training,all mice were carried out exhaustive experiments to test endurance capacity of exercise.Y-maze test was performed 24 h later to test the ability of learning and memory.The expression of autophagy related proteins LC3,p62,Parkin and AMPK/AKT/mTOR signaling pathway proteins were detected by Western blot.Results:1 The effect of exercise,trehalose and exercise combined with trehalose on exercise tolerance in aged mice:Compared with the mice of YC group,the exhaustive time of OC group was significantly decreased(P<0.01);Compared with the OC group mice,the exhaustive time of T group mice was no significant difference,the exhaustive time of E group and E+T group ware significantly prolonged(P<0.05,P<0.01);Compared with T group,the exhaustive time of E group and E+T group ware significantly prolonged(P<0.05,P<0.01).Compared with E group mice,E+T group mice’s exhaustive time was also significantly prolonged(P<0.05).2 The effect of exercise,trehalose,and exercise combined with trehalose on learning and memory ability in aged mice:Compared with the YC group mice,the time of finding safe area of OC group mice was significantly prolonged(P<0.01),the time of finding safe area of T group,E group and E+T group were significantly decreased(P<0.05,P<0.05,P<0.05);Compared with the OC group,the time of finding the safe area in the T group,E group and E+T group were significantly decreased(P<0.05,P<0.05,P<0.01).3 The effect of exercise,trehalose and exercise combined with trehalose on cortex autophagy signal pathway in aged mice:Compared with YC group,the expression level of p62 protein in the other groups did not change significantly.Compared with YC group,the ratio of LC3-Ⅱ/LC3-Ⅰin OC group was significantly decreased(P<0.05);Compared with OC group,the expression of the ratio of LC3-Ⅱ/LC3-Ⅰin T group,E group and E+T group were significantly increased(P<0.01,P<0.01,P<0.01).Compared with YC group,the expression level of the LC3-Ⅱprotein in OC group was significantly decreased(P<0.05);Compared with OC group,the expression level of the LC3-Ⅱprotein in T group,E group and E+T group were significantly increased(P<0.01,P<0.01,P<0.01).Compared with the YC group,the expression level of Parkin protein in OC group was not significantly difference(P<0.05),the expression level of Parkin protein in E group and E+T group were significantly increased(P<0.05,P<0.05);Compared with the OC group,the expression level of Parkin protein in T group,E group and E+T group were significantly increased(P<0.05,P<0.01).Compared with the YC group,the expression level of Bnip3 L protein in the OC group was significantly decreased(P<0.05);Compared with the OC group,the expression level of Bnip3 L protein in T group,E group and E+T group were significantly increased(P < 0.05,P<0.05,P<0.01).(2)Effects of exercise on AMPK/AKT/mTOR signaling pathway in aged cortexCompared with the YC group,p-AMPK/AMPK in the OC group was significantly decreased(P<0.05);Compared with the OC group,p-AMPK/AMPK in T group,E group and E+T group were significantly increased(P<0.05,P<0.05,P<0.05).Compared with the YC group,p-mTOR/mTOR in the OC group was no significantly difference;Compared with the OC group,p-mTOR/mTOR in T group was no significantly difference,E group and E+T group were significantly decreased(P<0.01,P<0.05).Compared with the YC group,p-S6k/S6 k in the OC group was no significantly difference;Compared with the OC group,p-S6k/S6 k in T group was no significantly difference,E group and E+T group were significantly increased(P<0.01,P<0.05);Compared with T group,p-S6k/S6 k in group E was significantly decreased(P<0.05).Conclusions:1.Exercise activated cortex autophagy signaling pathway in aged mice through AMPK/mTOR signaling pathway,improved the cognitive function.2.Trehalose activated cortex autophagy signaling pathway in aged mice through mTOR independent signaling pathway,improved the cognitive function. |
