Objectives:we will discuss the anti-pulmonary fibrosis effects and possible mechanisms of hydrogen sulfide by observing the expression of HO-1 and TGF-? 1 in rats with pulmonary fibrosis.Methods:Sixty 6-week-old healthy male SD rats were choosed and randomlyseparated into four groups:control group(group A),bleomycin group(group B),bleomycin +prednisone acetate group(group C),bleomycin + sodium hydrosulfide(the H2S donor)group(group D).Bleomycin was intratracheally administrated at a dose of 5mg/kg for once making the model of pulmonary fibrosis.But equi-valent volume of normal saline was given in groupA by the same way.The first day after eastablishing models,rats of groupD were adiministered intraperitoneal injections of NaHS(28 ? mol/kg)everyday,prednisone acetate(0.56mg/kg)was taked by means of intragastric administration everyday to the rats in groupC.The equal volume of normal saline was administrated by means of intragastric administration in group A and B.5 rats in each group were sacrificed randomly on the day7,14 and 28.The left lung tissue were collected for HE stain and Masson stain and for the expressions of the protein of HO-1 and TGF-? 1 analysed by means of immunohistochemistry(PV);The right lung tissue were collected for the expressions of HO-1 and TGF-?1 mRNA by means of RT-PCR respectively.Results:1?HE and Masson staining:On the 7th day,compared with groupA,the alveolar structure was disrupted,instead,a large number of inflammatory cells infiltrated into alveolar space and pulmonary interstitium and collagen fiber deposition was seen in some areas in group B.On the 14th day,the alveolar structure was further disrupted,more inflammatory cells and pulmonary interstitium increased.On the 28th day,the lung tissue was completely disordered,a large number of collagen fibers formed instead.Similar changes occurred in groupC and groupD,but the degree of fibrosis was weaked than groupB and aggravated than groupA.2?MDA content:Compared with group B,C,D,the content of MDA in group A reduced,the differences above were siyiticant(P<0.05).Compared with group B,the content of MDA in group C,D reduced,but increased compared with the group A.The differences above were siyiticant(P<0.05)3?Immunohistochemistry(PV)and RT-PCR results:(1)Compared with group B,C,D,the level of protein of TGF-? 1 and HO-1 in group A reduced,the differences above were siyiticant(P<0.05).Compared with group B,the level of protein of TGF-? 1 and HO-1 in group C,D reduced,but increased compared with the group A.The differences above were siyiticant(P<0.05).(2)Compared with group B,C,D the level of mRNA of TGF-? 1 and HO-1 in group A reduced,the differences above were siyiticant(P<0.05).Compared with group B,the level of mRNA of TGF-? 1 and HO-1 in group C,D reduced,but increased compared with the group A.The differences above were siyiticant(P<0.05).Conclussions:Exogenous hydrogen sulfide can delay the process of bleomycin-induced pulmonary fibrosis in rats.The mechanism of exogenous hydrogen sulfide in resistance to pulmonary fibrosis may regulate oxidative antioxidant imbalance,inhibite the expression of TGF-? 1 and down regulation the.expression of HO-1.
