Objective: This study was aimed at the construction of an attenuated and highly immunogenic vaccinia virus vector.Methods: Constrct a TK2L-TC7 L deleted vaccinia virus Tiantan strain by homologous recombination technique,and vaccinia virus TK / TC VTT was screened by EGFP and Cre / Lox P recombinant system.In vitro experiments,vaccinia virus TK / TC VTT and VTT were analyzed for diffusion and cytotoxicity by crystal violet staining and MTS detection.In vivo experiments,virulence of the gene-deficient virus TK / TC VTT was analyzed by changes of body weight and mortality in mice;and the specific antibody level and T cell proliferation were used to analyze the immunogenicity of TK / TC VTT.Results: Deletion of TK2L-TC7 L gene deletion did not affect the normal replication of the virus,and significantly attenuated the virulence of the virus and effectively induced the immune response.Conclusion: This study provides an attenuated and efficient vaccinia virus vector vaccine for future medical fields.
