| Objective: this experiment using the method of serum pharmacology,the expression of ILK in podocyte of Jiedutongluobaoshen serum induced by in vitro high glucose,affect the regulation of PI3K/Akt signaling pathway and related factors on the occurrence of apoptosis,the mechanism of cell injury of foot Jiedutongluobaoshen medicine intervention in high glucose conditions,further to provide the experimental basis for Jiedutongluo Paul the kidney on DN theory.Methods: to establish the model of podocytes induced by high glucose.Podocyte in vitro model + drug intervention factors of.A groups: normal glucose group;B group: high glucose group;C group: high glucose + low dose drug serum group;D group: high glucose +middle dose drug serum group;E group: high glucose + high dose drug serum group.The detection of ILK,enzyme-linked immunosorbent assay Akt,Bax,Bcl-2,Caspase-3 expression level.The detection rate of podocyte apoptosis by flow cytometry.Results: experiment one: by enzyme linked immunosorbent assay results showed that:compared with group A,B,C,D,E group,the expression of ILK was significantly increased(P < 0.05),compared with group B,C,D,ILK decreased expression of E group,C group had no statistical significance(P > 0.05),D,E group was statistically significant(P < 0.05);compared with group A,B,C,D,Akt expression in E group were lower(P < 0.05),and compared with group B,C,D,E group increased the expression of Akt,E group was statistically significant(P<0.05),C,D group no statistical significance(P > 0.05).Experiment two: compared with group A,B,C,D,E group Bax,the expression of Caspase-3 was significantly increased(P<0.05),compared with group B,C,D,Bax decreased expression of E group,C group had no statistical significance(P > 0.05),D,E group was statistically significant(P < 0.05),C,D were lower in E group the expression of Caspase-3(P<0.05);compared with group A,B,C,D,E and Bcl-2 expression was decreased(P<0.05),and compared with group B,C,D,E group increased the expression of Bcl-2,C group had no statistical significance(P > 0.05)D,E group was statistically significant(P < 0.05).Flow cytometry and podocyte apoptosis results showed that: compared with group A,B,C,D,apoptosis rate of E group was increased(P<0.01),compared with group B,C,D,E groupapoptosis rate(P<0.01)decreased,compared with the C group,D group,E group apoptosis rate reduce(P<0.01).Conclusion: 1.The protective mechanism of Jiedutongluobaoshen on glucose stimulated podocyte injury probably by blocking the PI3K/Akt signaling pathway mediated by ILK activation,inhibition of Bax and Caspase-3,increase the expression of Bcl-2,reduce podocyte apoptosis and play a role in.2,indicated that the ILK-PI3K/AKt signal transduction pathway is a key.3 high glucose induced podocyte apoptosis,to provide the experimental basis for the treatment of renal DN Jiedutongluo protection law. |
