Objective To investigate the expression level of PRDX1 in non-small cell lung cancer(NSCLC),and analyze the relationship between PRDX1 expression and various clinicopathological characters.This study aimed to evaluate the role of PRDX1 in early diagnosis,metastasis and prognosis of NSCLC,and possibility for PRDX1 as a new target in cancer therapy.Methods 1,Seventy-two NSCLC specimens and corresponding normal lung tissue with complete follow-up data were enrolled in the study,including patients' clinicopathological characters such as age,sex,clinical stage and overall survival time.2,Tissue microarrays and immunochemical(IHC)staining was performed to detect the expression levels of PRDX1 in normal lung tissue and NSCLC tissue.3,The relationship between PRDX1 expression and various clinicopathological characters was evaluated.Kaplan-Meier analysis was performed to explore the relationship between PRDX1 expression and overall survival of the patients.4,Querying The Cancer Genomic Atlas(TCGA)database to analyze the relationship among PRDX1 expression,various clinicopathological characters and survival time.Results 1,The IHC staining showed that the expression of PRDX1 was presented both in NSCLC tissue and normal lung tissue and mainly located in cytoplasm.2,The expression level of PRDX1 was significantly increased in NSCLC when compared with the normal lung tissue.High expression rate of PRDX1 in NSCLC and normal lung tissue were 75.0%and 26.2%,respectively.(P<0.05)3,High expression of PRDX1 was correlated with lymph node metastasis(P=0.031),advanced TNM stages(P=0.037)and worse differentiation status(P=0.023)in NSCLC,and uncorrelated with age,gender and tumor size(all P>0.05).4,Kaplan-Meier analysis showed that PRDX1high patients suffered significant worse(P=0.048)overall survival rate than PRDX1low patients in NSCLC.Median overall survival in PRDXl1ow group(64.3(95%CI 48.0-80.6)months)was obviously longer than that in the PRDX1high group(49.6(95%CI 38.5-60.7)months).5,According to the TCGA database,the mean intensity of PRDX1 in NSCLC vs normal alveoli were 0.24 ± 0.74 vs-0.20 ± 0.41(P<0.001).Kaplan-Meier analysis showed that PRDX1high patients suffered significant worse(P=0.035)overall survival rate and progression-free survival rate(P<0.001)than PRDX1low patients in NSCLC.Conclusion 1,The expression level of PRDX1 was significantly increased in NSCLC tissue when compared with the normal lung tissue.2,Elevated expression of PRDX1 was correlated with lymph node metastasis,advanced TNM stages and worse differentiation status in NSCLC.3,Elevated expression of PRDX1 was correlated with worse prognosis.Therefore,PRDX1 could be regarded as an important indicator of prognosis in NSCLC.4,According to the TCGA database,elevated expression of PRDX1 was correlated with lymph node&distant metastasis,advanced TNM stages and worse prognosis in NSCLC.
