Objective:To explore the effect of Salidroside(Sa)in protecting skeletal muscle in ratswith chronic obstructive pulmonary disease(COPD)and the relationship withPI3K/Akt/mTOR.Method:40 adult SD rats were divided into 2 groups,the smoking group of 24,the blank group(C)of 16.The smoking group adopts pure passive smoking to establishsmoking rat model,whlie the blank group take false exposure method to establish the model of the blank group.Four mouth later,each group take 8 rats to evaluate the COPD rat model.Then the smoking group were divided into 2 groups,COPD model group(CM),Salidrosideintervention group(Sa),each group of 8.Salidrosideintervention group(Sa)give a dose 25 mg / 100 gsalidroside lavage treatment twice a day last 1 month.While Blank group(C)and COPD model group(CM)given a same dose of normal salion for 1 month.Gather lung tissue and quadriceps,lung tissue pathology slice,The protein contents of phosphorylation and total PI3 K,Akt,mTOR,4E-BP1,p70s6 k in skeletal muscle were detected by Western blot,theprotein expression of phosphorylation and total PI3 K,Akt,mTOR,4E-BP1,p70s6 k in skeletal muscle were detected by immunohistochemistry.Results:1.Western blot protein expression comparison:(1)p-PI3K:group C(0.39±0.04),the CM group(0.25±0.01),Sa group(0.32±0.02);(2)PI3K:group C(0.61±0.04),the CM group(0.47±0.03),Sa group(0.54±0.01);(3)p-Akt:group C(0.41±0.04),the CM group(0.15±0.03),Sa group(0.24±0.03);(4)Akt:group C(0.55±0.04),the CM group(0.41±0.02),Sa group(0.48±0.03);(5)p-mTOR:group C(0.45±0.03),the CM group(0.27±0.01),Sa group(0.35±0.02);(6)mTOR:group C(0.55±0.04),the CM group(0.43±0.02),Sa group(0.49±0.02);(7)p-4E-BP1:group C(0.47±0.04),the CM group(0.25±0.04),Sa group(0.39±0.02);(8)4E-BP1:group C(0.57±0.05),the CM group(0.41±0.02),Sa group(0.49±0.01);(9)p-P70S6K:group C(0.44±0.04),the CM group(0.27±0.04),Sa group(0.32±0.02);(10)P70S6K:group C(0.52±0.05),the CM group(0.35±0.04),Sa group(0.44±0.02);Compared with group C,the CM group decreased phosphorylation and total PI3 K,Akt,mTOR,p70s6 k,4E-BP1 expression,and the value between the difference had statistical significance(P < 0.05).Sa group compared with group CM,increased phosphorylation and total PI3 K,Akt,mTOR,p70s6 k,4E-BP1 expression,and the value between the difference had statistical significance(P < 0.05).2.Immunohistochemistry result:(1)p-PI3K:group C(0.106±0.009),the CM group(0.048±0.007),Sa group(0.077±0.004);(2)PI3K:group C(0.155±0.005),the CM group(0.061±0.002),Sa group(0.114±0.008);(3)p-Akt:group C(0.095±0.003),the CM group(0.041±0.004),Sa group(0.071±0.004);(4)Akt:group C(0.121±0.006),the CM group(0.053±0.004),Sa group(0.088±0.008);(5)p-mTOR:group C(0.099±0.008),the CM group(0.032±0.003),Sa group(0.063±0.005);(6)mTOR:group C(0.121±0.004),the CM group(0.069±0.005),Sa group(0.088±0.005);(7)p-4E-BP1:group C(0.086±0.009),the CM group(0.031±0.001),Sa group(0.058±0.002);(8)4E-BP1:group C(0.110±0.007),the CM group(0.051±0.002),Sa group(0.086±0.004);(9)p-P70S6K:group C(0.099±0.003),the CM group(0.041±0.002),Sa group(0.070±0.007);(10)P70S6K:group C(0.118±0.003),the CM group(0.067±0.005),Sa group(0.095±0.003);Compared with group C,the CM group decreased phosphorylation and total PI3 K,Akt,mTOR,p70s6 k,4E-BP1 expression,and the value between the difference had statistical significance(P < 0.05).Sa group compared with group CM,increased phosphorylation and total PI3 K,Akt,mTOR,p70s6 k,4E-BP1 expression,and the value between the difference had statistical significance(P < 0.05).Conclusion:Salidrosidemay be through the PI3K/Akt/mTOR signal pathway to protect muscle atrophy in rats with COPD.
