| Objective:Understanding of bone marrow MSCs on radioactive damage in colorectal tissue of rats radioactive damage repair and the influence of cell apoptosis and related gene differential expressionMethods:36 healthy female SD rats,were randomly divided into 3groups(blank control group,gamma irradiation group,BMSCs treatment group),each group of 12.A blank control group: no light,tail intravenous saline experiment 2 days 1 ml;Gamma irradiation groups: experiment 1 day one-time full abdomen in 60 co gamma irradiation(total dose of 12 gy),tail intravenous saline experiment 2 days 1 ml;MSCs treatment groups:experiment 1 day with 60 co gamma rays for one-off full abdomen irradiation,12 gy total dose,experiment 2 days between tail vein injection of rat bone marrow mesenchymal stem cells(RBCMSCs)injection of 1 ml(1 x106 / ml).Three groups of rats in the seventh day of experiment executed and colorectal,small intestinal mucosa pathological morphological changes under light microscopy observation.By using in situ end labeling(TUNEL)to detect colorectal mucosa cell apoptosis.ELISA method for determination of peripheral blood IL-10,TNF-a content,in order to understand theintestinal inflammatory reaction.The application of gene chip technology to the level of mRNA expression in the three groups of rats in colorectal comparative hybridization analysis,to explore the differentially expressed genes and statistical analysis.Results:1.Colorectal tissue HE staining: gamma irradiation rats colorectal mucosa morphology and structure compared with blank control group had obvious changes,intestinal mucosal thickness uneven,fluffy structure is incomplete,submucosal edema,cavitation appearance change,intestinal crypt lost normal structure,submucosa visible vasodilation,blisters,inflammatory cells infiltration;Compared with BMSCs treatment group,BMSCs treatment group colorectal mucosa pathological damage in rats.2.Colorectal mucosa apoptosis rate: gamma irradiation rats colorectal mucosa apoptosis rate compared with blank control group were significantly higher(P < 0.05),and the apoptosis rate of BMSCs treatment group are higher than gamma irradiation group(P < 0.05).3.Peripheral blood content of IL-10 : the level of IL-10 in the irradiation group rats peripheral blood were compared with blank control group were significantly lower(P < 0.05),and IL-10 levels of BMSCs treatment group were higher than gamma irradiation group(P < 0.05).4.Peripheral inflammatory factor TNF-a content: In the peripheral blood of the gamma irradiation group were compared with blank controlgroup were significantly higher(P < 0.05),whereas TNF-a content in BMSCs treatment group were lower than gamma irradiation group(P <0.05).5.Application of gene chip technology in blank control group,gamma irradiation group,the BMSCs RNA in the treatment group rats colorectal,comparing on DRA expression level hybridization analysis: find the analyzed so gene in the three groups of both two comparison suggest a variety of genetic variability is an expression,which contains multiple genes of rise and fall.These gene expression products related to cell apoptosis,signal transduction,cell adhesion,enzyme activity,immune response,receptor regulate every aspect,and gastrointestinal tract inflammation,gastrointestinal tract,colorectal cancer,leukemia,radioactive damage,colorectal cancer,liver cancer,lung cancer,cervical cancer,breast cancer and other disease development have significant correlation.Conclusion:1.BMSCs transplantation BMSCs transplantation alleviate radioactive colorectal lesions in rats.2.BMSCs transplantation may via reducing apoptosis rate to alleviate radioactive colorectal lesions in rats.3.BMSCs transplantation could inhibit inflammation to reduce radioactive colorectal lesions in rats.4.BMSCs transplantation able to influence from gene level radioactive colorectal lesions in rats.5.BMSCs transplantation cure radioactive colorectal lesions in rats through down regulating the expression of CCR5. |
