Aim: The purpose of this study is to measure the thickness of the palate mucous and analysis the thickness of palatal mucosa in association with age and gender in order to provide a theoretical reference for the clinicians to choose the appropriate location, method, size and thickness when get connective tissue graft.Materials and Methods: Sixty healthy subjects were included under two groups(aged 18-29 years and 40-59 years),each group consisted of 15 males and females.Under lidocaine(local anaesthesia)and using Root canal file measure 15 different sites on palatal mucosa, then measure the thickness of palant mucosa at different age and sex. Finally, statistics and analysis of the correlation between eachindex. Results: The palate mucosa gradually thickness with the distance from the gingival margin. The average thickness at the 2mm,7mm and 10 mm of all teeth were2.53 mm,3,44 mm and 4.01mm;From the canine to 2nd molar average thickness are2.43±0.35mm?2.71±0.45mm?3.38±0.46mm?3.36±0.58mm?3.83±0.56 mm.And at the 2,7,10 mm points except 1st molar, the thickness of the palate mucosa is gradually thickening. At the site of 7mm,the 2nd molar is the thickest and 1st molar is the thinnest. The thickness of the palate mucous in the middle and old age group is greater than the young group(p<0.05).And in the male of middle and old age group is greater than the female, there is a significant statistical significance(p<0.001).No statistical significance between male and female in the youth group of the palate thickness(p>0.05).Conclusion: Palatal masticatory mucosa is thicker in older age group than in younger age group and it is thinner in females as compared to than in males. The palate mucosa gradually thickness with the distance from the gingival margin.The2 nd permolar to mid palatal palatal aspect of the 2nd molar are the appropriate donor site for the acquisition of connective tissue graft and this area can get at least 1mm Subepithelial connective tissue graft.
