Background Fracture is the most common disease in orthopedic clinical practice.With the increase of complicated fractures caused by high energy damage such as traffic accidents,the incidence of delayed union or even nonunion of fractures is obviously increased,which causes great pain to the patients' physical and mental.How to effectively and effectively promote fracture healing has always been a hot topic in the field of orthopedics.Local blood vessel formation and stable blood supply are the prerequisites for normal healing of fractures,and their generation and reconstruction provide a powerful microenvironment for local bone regeneration.Vascular endothelial growth factor / vascular endothelial growth factor receptor(VEGF / VEGFR)signaling pathway has a key regulatory effect on neovascularization.Objective Taohong Siwu Decoction is the representative agent of traditional Chinese medicine "Huoxue Huayu,Quyu new" theory,treatment of fractures and bone defects.It has the functions of supplementing blood and promoting blood circulation and removing blood stasis.In recent years,it has been found that the Taohong Siwu Decoction The expression of VEGF in the callus tissue of the fractures was stimulated by the proliferation of the callus neonatal vessels in the early stage of the callus,and the microvascular remodeling and microcirculation reconstruction in the callus of the fracture were accelerated.However,the expression of VEGF protein,Callus microvascular neonatal effective intervention of the molecular biologica l mechanism to be further studied.Methods 64 healthy healthy rats were randomly divided into two groups: the placebo group and the placebo group(200.0 ± 40)g(Henan province animal experiment center).The rats were randomly divided into two groups: the placebo group 32 rats were treated with Taohong Siwu treatment group,and the tibial fractures were given at the same time.The two groups were given gavage once a day.The treatment group was treated with Taohong Siwu Decoction and the same amount of dist illed water.Thecontrol group was the same amount of distilled water.The animal s were sacrificed at 7,14,21 and 28 days after operation.Eight rats were sacrificed at each time.The local callus and surrounding tissues were sacrificed.Immunohistochemi cal treatment was performed to observe the expression of VEGF.HE staining was used to observe the microvessels newborn.Results The histological findings of callus showed that a large number of granulation tissues were formed in the experimental group and the control group.The microvessel formation was more in the experimental group than in the experimental group.On the 14 th day after operation,the number of blood vessels in the control group was slightly higher than that in the control group(P <0.05).On the 21 st day after operation,the number of microvessels in the callus tissue decreased,and the number of microvessels in the callus increased compared with the 7th day after operation,but still less than the experimental group.On the 28 th day after operation,the number of microvessels in the callus tissue of the experimental group and the control group decreased.Immunohistochemistry showed that a certain number of VEGF-positive cells were observed in the experimental group on the 7th day after operation.A small number of positive cells were seen in the control group.On the 14 th day after operation,the number of positive cells in the experimental group reached the peak value,while the positive cells in the control group also reached the peak but less than the experimental group.On the 21 st day and the 28 th day after operation,the number of positive cells in the experimental group and the control group decreased gradually,but the number of positive cells in the experimental group was higher than that in the control group.Compared with the control group,there were significant differences in the number of VEGF positive cells and the number of microvessel in the callus tissue at each time point in the experimental group.And the number of VEGF positive cells and the number of microvascular neonates were significantly correlated with the application of Taohong Siwu Decoction.The number of VEGF positive cells in the callus tissues of the two groups was similar to that of the two groups.The peak period was the same on the 14 th day after operation.While the number of microvessel in the callus tissue of the experimental group was higher than that of the control group.Conclusion: Taohong Siwu Decoction can significantly promote the expression of VEGF in local callus of fractures,and stimulate the expression of VEGF and microvessel in the local microvascular neonates.The expression of VEGF and microvessel can be maintained at a high level and promote fracture healing.The peak of microvascular neonatal tissue in callus tissue was advanced,but it did not affect the peak time of VEGF expression.
