Colorectal cancer(CRC)is common cancer with high mortality worldwide,its survive rate is relative with the stage when been diagnosed and treated:the patients who are detected and treated at early stage have greater oppotunity to survive.Colonoscopy is still the gold standard in detecting CRC,but not optimal for screen because of its invasiveness and demand of anesthesia.Tumor biomakers is noninvasive and economical,but the sensitivity and specificity of curecent commonly used biomarkers for early stage CRC are still not ideal,searching for new biomarkers is urgely needed.Purpose:To evaluate two new tumor biomarkers RBP4 and THBS2 for diagnosis and prognosis of CRC,analyze their advantage and disadvantage compared with the commonly used biomarkers CEA and CA19-9.METHODS:Detect serum concentration of RBP4 and THBS2 from 402 CRC patiants and 218 controls.Statistical analysis was carried out using the SPSS 22.0 software package.Univariate comparisons between groups(cases and controls)were performed using chi-square tests or Fisher exact tests for categorical data,and using Mann-Whitney U test for continuous variables.A receiver operating characteristic(ROC)curve was plotted to assess diagnostic performance.Results:The results showed that the average RBP4 and THBS2 concentrations in normal controls were significantly lower than in CRC patients(36.5±11.4μg/mL vs 21.8±8.7μg/mL and 20.5±6.1ng/mL vs 14.5±7.3ng/mL,respectively),both p<0.001.RBP4 distinguished CRC patients from normal individuals with the area under the receiver operating characteristiccurve(AUC)performing at 0.852,with sensitivity of 74.9%and specificity of 81.7%.While THBS2 distinguished CRC patients performing AUC at 0.794,with sensitivity of 64.9%and specificity of 87.1%.The ability of RBP4 and THBS2 serum concentration distinguishing CRC from normal controls showed better than that of serum CEA(AUC=0.818)or CA-199(AUC=0.650)concentration.Conclusions:The potential role of serum RBP4 and THBS2 as biomarker in colorectal cancer(CRC)diagnosis has never been studied.This is the first study to report RBP4 and THBS2 as diagnosis serum biomarkers for CRC,finding that the performance of RBP4 and THBS2 such as sensitivity and specificity especially for early stage CRC showed better than that of serum CEA or CA-199.which might be a good supplement for CEA or CA19-9 for clinical diagnosis.
