Telomerase is an RNA-protein complex that is present in eukaryotic cells and has the effect of prolonging the telomere sequence at the end of the intracellular chromosome.The length of the telomere sequence is closely related to the cell division ability.Since the telomere sequence is shortened by the action of the replication mechanism,the telomere sequence will be lost when it is reduced to a certain extent.However,in the cancer cells,the activity of granzyme is high,and the length of telomere sequence can be prolonged.Therefore,telomerase is considered to be an important reason for the immortalization of cancer cells.In recent years,telomerase has been used as an important marker in cancer research and has been used in a large number of applications,including cancer diagnosis and cancer treatment.The activity of telomerase is directly related to the ability of cancer cells to divide,so a large number of studies on telomerase activity testing have been reported.Among them,the fluorescence sensor because of its fast response,simple design,efficiency,selectivity and other advantages,is widely used in the detection of telomerase activity.However,monochromatic fluorescent probes have the effect of preventing false positives from interfering with environmental factors.Therefore,this paper designed a multi-color multi-color ratiometricfluorescent DNA probe,can effectively detect telomerase activity and can avoid the emergence of false-positive signals at the same time.The fluorescent probe could show different colors according to the level of the telomerase activity,so we can identify several different cancer cell and normal cell with naked eye under the UV light.The first chapeter is divided into two parts.The first part mainly introduces the telomere sequence and the structure and function of telomerase.At the same time,the earliest methods of detecting telomerase activity,as well as the new and classic methods reported in recent years,have also been analyzed for their advantages and disadvantages in terms of the principle of each method.The second part mainly introduces several types of classical fluorescent probe types,analyzes its luminous principle,and illustrates the structure and application of various probe typesIn the second chapter,we mainly introduce the principle of multi-color ratio fluorescent DNA probe to detect telomerase activity in cell extracts.By modifying the red,green fluorescent groups and quenching groups of DNA ends,and the targeting of DNA sequences Design,the use of telomerase can be extended to extend the telomerase-induced sequence and base complementary pairing principle,to achieve red and green fluorescence to open and close.At the same time,the feasibility of the protocol was analyzed,and the ability of the probe to detect telomerase activity in cancer cells was verified,and the telomerase activity in the cell extracts of various cancer cells was compared.The effect of telomerase inhibitor on telomerase activity was also investigated by our probe.
