Experimental Study On The Intervening Effects Of HuoTan JieDu TongLuo Extract On Acute Myocardial Ischemia Model In Rat

Purpose: In this study,the rat model of acute myocardial ischemia was induced by intraperitoneal injection of isoproterenol hydrochloride.To discuss and analyse theeffects of HuoTan JieDu TongLuo extract on the treatment and protection of acute myocardial ischemia rats,and to lay the foundation for further study,the studyobserves the effects of different doses of HuoTan JieDu TongLuo extract on ST-segmentation of electrocardiogram II lead,serum myocardial enzymes(CK,LDH,AST),troponin I(Tn-I)and ischemic modified albumin(IMA),superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)(MDA)content and nitric oxide(NO)content in the experimental rats.Method: The rats were randomly divided into 5 groups,the model group,the positive control drug Tongxinluo capsule group,the HuoTan JieDu TongLuo extract high dose group,the HuoTan JieDu TongLuo extract middle dose group and the HuoTan JieDu TongLuo extract in the low dose group,with 10 rats in each group,givingcorresponding drugs once a day for 14 daysrespectively.Rats in the model group were given equal volume of distilled water.From the 12 th day,rats in each group were treated with isoproterenol 20 mg / kg / s by intraperitoneal injection.The injection is givenonce a day,continuously for 3 days.After the last administration of the test drug for 30 min,the standard II electrocardiogram is recorded and the isoproterenol 20 mg / kg is injected.Record the standard II electrocardiogramafter the injection of isoproterenol at 10 min,20 min and 30 min.After the observation take the abdominal aorta blood sample and weighquantitative heart.Calculate the electrocardiogram(ECG)ST segment offset mV at 10 min,20min and 30 min after myocardial ischemia of the rats,and detect the activities of CK,LDH,AST,the content of cTn-I and IMA,the activities of SOD,GSH-Px,and the content of MDA and NO.Result: It is possible for HuoTan JieDu TongLuo extract to significantly reduce the ECG ST segment offset,the activity of serum CK,LDH and AST three enzymes in acute myocardial ischemia rats and the content of myocardial injury markers cTn-I and IMA to improve themyocardial ischemic degree of cardiomyocytes in acute myocardial ischemia rats;It can significantly increase the activity of myocardial SOD and GSH-Px,reduce the content of MDA,and significantly increase the NO content,thus improve vascular endothelialinjury,improve rat antioxidant capacity of myocardial cells.Conclusion: HuoTan JieDu TongLuo extract can significantly prevent myocardial injury in acute myocardial ischemia ratsinduced by isoproterenol.The mechanism may be related to itsprevention of vascular endothelial damage andthe enhancement of antioxidant capacity of rat cardiomyocytes.
