Purpose:1.To compare the expression of LncRNA gene in four tumor tissues and adjacent tissues,and to obtain the high expression of LncRNA in tumor tissues,and then use qRT-PCR to verify the consistency of the expression level and high throughput sequencing results.It was found that the expression level of LncRNA was higher in the osteosarcoma tissues under the 2 methods.2.The purpose of this study was to investigate the effect of LncRNA on the biological characteristics of osteosarcoma cells.Method:This study first collected clinical specimens of 4 tumor of whole genome sequencing,comparative differences between cancer tissue and tumor LncRNA gene expression analysis of tumor tissue,high expression of both genes were 6 LncRNA,by using qRT-PCR to verify its expression level and high-throughput sequencing results.Bioinformatics analysis showed that ERVK13-1 was related to PI3K/Akt,AMPK,mTORC and so on.MG63 cells were used to study the effects of ERVK13-1 on the growth of tumor cells.qPCR was used to detect the protein expression of ERVK13-1 and downstream.Result:1.The expression level of ERVK13-1 in osteosarcoma was increased by 2 methods.So we choose ERVK13-1 as the research object.2.After CRISPR-cas9 knockdown of ERVK13-1 promoter,the proliferation ability of MG63 cells decreased and the apoptosis increased.3.Knockdown of ERVK13-1 significantly decreased the level of mRNA transcription and protein expression in Akt.Conclusion:ERVK13-1 enhances the function of PI3K-Akt signaling pathway by up regulating the expression of Akt,and promotes the survival of tumor cells and various malignant manifestations.The gene can be used as a marker of malignant degree of tumor and a target of intervention.
