[Objective]To detect fatty acid binding protein-5(FABP-5)gene expression in various human ovarian cancer cells,investigate the effect of interruption of FABP-5 gene on its expression by small interference RNA(siRNA)and explore its effect on the proliferation,migration,apoptosis in the ovarian cancer cells[Methods]The real-time PCR was explored to detect FABP-5 expression in various human ovarian cancer cells.The FABP-5 gene siRNA interference fragment(si-FABP-5)and siRNA negative control(si-Con)were respectively designed and transfected into SKOV3 cells which has the highest FABP-5 relative expression among SKOV3,A2780,8910,OVCAR3 four different ovarian cancer cells.Real-time PCR were used to detect the interference efficiency of FABP-5 in SKOV3 cells transfected with si-FABP-5 and si-Con.Protein expressions of FABP-5 was detected by western blot assay,respectively.Proliferation related factors MKI 67 were also detected in two groups.Cell counting kit-8(CCK-8)assay was used to assess the proliferative activity of two groups of cells.Cell migration ability was measured by transwell experiment.The apoptosis rates of cells was assessed by the flow cytometry.[Results] The m RNA expression of FABP-5 gene in human ovarian cancer 8910 cells represented the lower relative expression and SKOV3 had a more increasing expression.After transfected transiently with si-FABP-5 and si-Con in SKOV3 cells,little obvious alterations of cellular condition were observed under light microscope.Compared with si-Con group,the proliferative activity of si-FABP-5 group cells at 24,48,72 hours were significantly reduced(P <0.01),along with the decreasing expression of proliferation-related factors MKI 67(P <0.05).Transwell migration assay showed that the penetrated cell counts of si-FABP-5 group was apparently lower than those in si-Con group.The flow cytometry results showed that the difference of apoptosis rate between si-FABP-5and si-Con group cells was not statistically significant(P> 0.05).[Conclusion] Our results suggested different human ovarian cancer cells existeddiverse expression levels of FABP-5 gene,and compared with other ovarian cancer cells,SKVO3 cells presented relative higher FABP-5 expression.Interruption of FABP-5 gene by siRNA effectively inhibited the mRNA expression of FABP-5 in SKOV3 cells,obviously reduced cell proliferative and migration activity,but not greatly affected the apoptotic rate.
