Objective:To investigate the potential role of hydrogen sulfide in the pathogenesis of endometriosis.Methods:?1?Ectopic endometrial tissues obtained from women with endometriosis?n=17?and eutopic endometrial tissue from women without endometriosis?n=30?were stained for the expression of CBS and CSE.?2?The cellular expression of CBS and CSE in normal and ectopic endometrial stromal cells was detected by immunocytochemical staining.?3?CBS and CSE in the ectopic and normal endometrial stromal cells were recruited to study the proliferative effect and possible mechanism of H2S by treating primary cultured normal human endometrial stromal cells?NESCs?with hydrogen sulfide?NaHS??pyrrolidinedithiocarbamate?PDTC?.?4?The influence of prototypical CBS inhibitor aminooxyaceticacid?AOAA?,and the CSE inhibitors DL-propargylglyicine?PAG?on the effect of lesion size and weight in the mouse model of endometriosis were detected through the allograft endometriosis mouse model.Results:?1?The expression of CBS and CSE in etopic endometrial tissues obtained from women with endometriosis were significantly higher than eutopic endometrial tissue from women without endometriosis.?2?NaHS can stimulate the proliferation of NESCs in a certain concentration range,after treating NESCs with NaHS and PDTC,the proliferative effect was antagonized in dose dependent manner.?3?AOAA,PAG or co-treatment can significantly reduce the weight and number of endometriosis lesions.Conclusion:1.hydrogen sulfide may be involved in the pathogenesis ofendometriosis,CBS and CSE have a certain potential as markers for evaluation endometriosis.2.sodium hydrosulfide can promote proliferation of NESCs via NF-?B signaling pathway.3.AOAA,PAG or co-treatment was effective in animal model of endometriosis,which provides a new target for treating endometriosis.
