Objective:Survivin molecular beacons was used to detect the expression of Survivin mRNA in endometrial carcinoma cell lines and endometrial carcinoma,and to evaluate its clinical value in the diagnosis of endometrial carcinoma.Methods:1.First with Western blot-(protein imprinting method)and PCR(Polymerase Chain Reaction,Polymerase Chain Reaction)in the detection of endometrial cancer cells and Survivin protein Survivin mRNA expression,and normal endometrial epithelial cells of choose and employ persons to do comparison,in confirming does have the expression of Survivin in endometrial cancer cells;Then the expression of Survivin mRNA in the lining of endometrial cancer cells was measured by means of the Survivin molecule beacon,and the feasibility of this method was understood.In order to test the accuracy of the test results in 40 cases of uterine membrane cancer and 40 cases of normal endometrial tissue samples,the results were verified by using the "Survivin" and the "Western" blot.2.Comparing the test results of the Survivin molecular beacon and the Western blot,the difference between the two was evaluated.Results:1.Western-blot tested positive expression of Survivin protein in the endometrial cancer cells,and the normal uterine epithelial cells were negative.2.The PCR method detects the lining of the endometrial cancer cells,Ishikawa,HEC-1-A,which can be detected in the form of A Survivin mRNA expression,whereas in the normal endometrial epithelial cells,it is not detected by the expression of the "Survivin mRNA".3.Ishikawa,Survivin molecular beacon in endometrial cancer cells were observed in HEC-1-A strong fluorescent signal,while in normal human endometrial epithelial cells did not see the fluorescent signal.4.Survivin molecular beacon detection in patients with endometrial carcinoma tissue specimens and normal endometrial tissue results showed: in endometrial carcinoma specimens organization visible fluorescence signal expression,while in normal endometrial tissue only weak or no fluorescence expression,40 cases of specimens in the organization was to 28 cases,70% sensitivity and speciality rate 87.5%,positive predictive value of 84.8%,74.4%,negative predictive value about the index of 0.57.5.Western-blot detecting endometrial carcinoma tissue samples and found normal endometrium tissues,endometrial tissue samples of Survivin protein positive expression,and in the negative expression in normal endometrium tissues,40 cases were positive for 30 cases of specimens in the organization,and sensitivity of 75%,the specific degree of 82.5%,positive predictive value of 81%,76.7%,negative predictive value about the index of 0.57.6.The differences in sensitivity and specificity were statistically significant(P > 0.05),in the expression of Survivin in the case of the Survivin molecular beacon and the Western blot of the endothelial cancer tissue specimen.Conclusion:1.The Survivin molecule beacon can detect the expression of the Survivin mRNA in the lining of the lining of the uterus.2.The Survivin molecule beacon is used to diagnose endometrial cancer with better sensitivity and specificity,which may be a viable,effective diagnostic method for diagnosing endometrial cancer.
