Objective To study the effects of PRDX1 protein expression on cell proliferation,invasion,migration and sensitivity of cisplatin in non-small cell lung cancer cells and reveal its related molecular mechanisms.Method Using shRNA technology to reduce the expression of PRDX1,the transfection efficiency was verified by QPCR,Western blot and immunofluorescence assay.Then proliferation,migration and invasion ability of lung cancer cells were detected by MTT,Wound Healing and Transwell assays,while confocal microscope was used to detect the intracellular localization of PRDX1 and the changes of the actin cytoskeleton.Western blot test the effect of PRDX1 on Akt?mTOR and ERK signaling pathway.Results the expression of PRDX1 in lung cancer cells was significantly higher than that in lung normal bronchial epithelial cells.The expression of cell lines in different lung cancer cells was different,A549,pc-9 and H1975 were significantly higher than 801D,95D and HCC827 cells.In the study of A549 cells,we found that PRDX1 was mainly located in the cell plasma and nucleus.PRDX1 Knockdown could significantly inhibit the A549 cells proliferation,migration and invasion ability and increase the sensitivity to cisplatin,meanwhile causing cell microfilament cytoskeleton rearrangement,withdecreased phosphorylation of Akt,mTOR and ERK.Conclusion Repression of PRDX1 gene expression can obviously reduce the lung cancer cell proliferation,migration,invasion ability and increase their sensitivity to cisplatin and its mechanism may be related to regulate cell microfilament cytoskeleton and Akt,mTOR and ERK pathway.
