Background: thyroid associated ophthalmopathy(thyroid associated ophthal,TAO)is a kind of orbital autoimmune disease which is closely related to thyroid disease(mopathy).With the basic research,especially the progress of immunology,further understanding of the etiology and pathogenesis of TAO are: TAO is composed of a variety of environmental factors(such as smoking),stress(such as trauma),genetic factors and other unknown function,stimulate immune function disorder.So far,none of the drugs in the treatment of TAO can achieve long-term efficacy without obvious side effects.Chinese medicine has obvious advantages in improving the symptoms of TAO.However,there are few studies on the mechanism of TAO in Chinese medicine,especially the lack of signal transduction in the study of TAO.Therefore,it is necessary to carry out the experimental research in this area.This experiment through the observation after CD40 L stimulation conflicts scattered on fibroblast series compound human retroocular(RFs)the effects of MMPs and TIMPs expression,so as to explore the mechanism of Chinese medicine in TAO,to explore the new method is effective and less side effects of long-term treatment of TAO.Objective: in this study,the clinical and experimental effective on the basis of cytokines from CD40 L on fibroblasts of human retroocular(RFs)the effects of MMP-1,MMP-2,TIMP-1,TIMP-2 expression,and explore the mechanism of purging fire conflicts scattered,blood conflicts scattered,raising the eye level process of powder in the treatment of TAO.Experimental methods and objects: 1 RFs were cultured and identified.Recovery of cryopreserved 2-8 generation RFs and observation of cell morphology.The effects of 2.CD40 L and three kinds of serum on the expression of MMP-1,MMP-2,TIMP-1 and TIMP-2 were determined by immunocytochemistry staining.Result:1 after successful in vitro culture of fibroblasts,the shape and shape of the cells were observed under light microscope.After 2-3 days,the cells were covered with 80%-90%.When the cells were identified by immunocytochemical staining,the results showed that the labeled cells were positive for vimentin(vimentin),and cytokeratin(cytokeration)was negative,which was determined to be RFs.2.CD40 L stimulation,the expression of MMP-1 and MMP-2 decreased,and the expression of TIMP-1 and TIMP-2 was up-regulated.After the intervention of the compound medicine serum,TIMPs could inhibit the expression of CD40 L,the expression of MMPs was up-regulated and the expression of TIMPs decreased.3.CD40 L stimulation,the ratio of MMPs/TIMP decreased compared with that before stimulation,and the ratio of MMPs/TIMP increased after the treatment of CD40 L.4.The best effect of restoring the balance of MMPs/TIMPs in the compound prescription of Ping Tu powder.Conclusion:CD40L stimulated human retroocular fibroblasts MMPs/TIMPs expression imbalance,traditional Chinese medicine containing serum can restore MMPs/TIMPs expression balance,fibroblast extracellular matrix deposition inhibition after CD40 L stimulation after the goal,to achieve the treatment of TAO.
