| Objective The purpose of this study is to explore the clinical significance of microRNA-4463,microRNA-6087 and micro RNA-4281 in the diagnosis,prognosis and clinical features of hepatocellular carcinoma,using the real-time PCR to detecting the expression of microRNA-4463,miRNA-6087 and mi RNA-4281 in the serum of normal human and patients with hepatocellular carcinoma.Methods Forty-five HCC patients blood were collected from December 2014 to December 2015,in the Nanhua Hospitaland and the First Affiliated Hospital Affiliated to the University of South China,and the clinical datas were collected.At the same time,the blood samples collected from 45 healthy people were collected from Health examination center of Nanhua Hospital Affiliated to the University of South China.And record the basic information.QRT-PCR was used to detect the expression of mi RNA-4281 、 miRNA-4463 and miRNA-6087 in the serum of healthy group and hepatocellular carcinoma patients.Results 1.Compared with the control group,the expression of miR-4463 and miR-4281 in serum of patients with hepatocellular carcinoma was significantly increased,the difference was statistically significant(P <0.05).But the expression of miR-6087 has not difference between normal and HCC patients.2.The expression level ofmiR-4463 in HCC which with low expression of AFP was significantly higher,even higher than that of high AFP group.The difference between the two groups was obvious,and statistically significant(P <0.05).And the expression of miRNA-4281 was not significantly correlated with AFP,and could be an independent diagnostic criteria for HCC.3.The area under the ROC curve was 0.805,and the sensitivity was 84.40%,and the specificity was 73.30% for microRNA-4281 in the diagnose of HCC.When combined with AFP,the area under the ROC curve and the sensitivity are increased,but the specificity is the same.4.There were no significant differences about the expression of miRNA-4281 and-4463 in different clinical characteristics such as gender,tumor size,stage of disease,liver function(transaminase,total bilirubin)and hepatitis B surface antigen of HCC(P> 0.05).However,the expression levels of miRNA-4281 and-4463 were divided into high and low groups.Kaplan-Meiermi analysis showed that the survival time of patients with high expression of RNA-4281 and-4463 was relatively short.The difference between the two groups was statistically significant(P <0.05).Conclusion 1.The expression of mi RNA-4463 and miRNA-4281 in serum of patients with hepatocellular carcinoma was significantly higher than that of normal controls,which can serve as a potential tumor marker for predicting liver cancer.2.Patients who had the high level of the expression of miRNA-4281 and miRNA-4463 result to a shorter duration of survival,and who had the low level live to a longer duration of survival.They are associated with prognosis and may serve as a novel marker for predicting prognosis.3.The expression of miRNA-4463 in HCC of who had the AFP negative in the serum is still significantly increased.So for AFP-negative patients,detecting mi RNA-4463 could help to diagnose HCC.The expression ofmiRNA-4281 was independent of the expression of AFP and could be used to help to diagnose HCC. |
