Object: The purpose of this subject is research on influence of the change of neurons in intestinal wall plexus including vasoactive intestinal peptide(VIP),nitric oxide synthase(NOS)and the gastrointestinal dynamics when the rats with severe acute pancreatitis(SAP).We are deeply discussed the mechanism of gastrointestinal dynamics changes leaded by severe acute pancreatitis in rat.These studies will provide certain theoretical basis for the research of clinical and basic with respect to salvia miltiorrhiza injection in severe acute pancreatitis.Method:60 SD rats were randomly divided into sham operation group(SO group)and severe acute pancreatitis(SAP group),each group include 30.After anesthesia laparotomy,only gently turn the duodenum and the pancreas in SO group,SAP group structure model of severe acute pancreatitis.Each rat place a piece of silver electrode in the pylorus proximal for Monitoring the stomach interdigestive myoelectric complex(IMC),every rat import saline according 2 ml / 100 g through the method of tail intravenous injection,every four hours.24 hours later Monitor IMC,Open the abdominal cavity again,rats were executed after observation of general situation of Abdominal viscera,whereafter,taking out pancreas,intestinal tissue for pathology examination,colon for detection of VIP NOS cexpression by double immunofluorescence.Result:(1)Each viscera in intra-abdominal: Obvious maroon ascites,pale yellow,saponification visible gastrointestinal congestion,gas effusion,and edema,large hemorrhage,necrosis,pancreas tissue dissolved has been found in the abdominal cavity of rats in SAP group.(2)double immunofluorescence test results of VIP,NOS neurons expression in Myenteric plexus of junction between the Knot bowel wall muscle :compared with SO group,colonic myenteric plexus NOS and VIP neurons expression were significantly increased in SAP group(P<0.01),a significant statistical significance.(3)each phase time of IMC and electric amplitude and frequency of IMC? phase of distal gastric smooth muscle : Cycle and ?,? phase time of IMC in SO group significantly shortened than SAP group(P<0.01),However,IMC?phase time,? phase peak amplitude and frequency highe rand long than SAP group(P<0.01)(4)small intestinal transit ratio :small intestinal transit ratio in SO group little than SAPgroup significantly(P<0.01);Statistical significance is obvious.(5)The pancreas,ileum pathology results: SO group no obvious pathological changes in light microscopic.large area of necrosis,inflammatory cells infiltration,most obvious broadening between lobes,acinar cell necrosis has been observed in SAP group.SO group rats ileum mucosa no obvious pathological changes,intestinal villus epithelial back gap obvious expansion,the top of the epithelium and lamina propria significant separation,lamina propria and visible capillary exposure,bleeding and ulcers can seeing in SAP group.(6)The pancreas,ileum pathological grading results: Compared with so group,SAP group significantly higher about pancreatic tissue pathology score(P < 0.01),ileal mucosal pathology score,(P<0.01).(7)Pearson correlation analysis results:In SAP group,Analysis found that the VIP,NOS neurons of knot bowel wall myenteric nerve plexus Positively correlated with the cycles of IMC(P<0.01),and IMC ? feng electric amplitude and frequency,small intestinal propulsion were negatively correlated(P<0.01)Conclusion:(1)The neurons of VIP and NOS reshaped in rat myenteric plexus of SAP rats.(2)The gastrointestinal motility weakene in rats with SAP rats?(3)There is some relationship may between gastrointestinal motility discrease with VIP,NOS neurons high expressive reshape in MP of colon wall of SAP rats.
