Purpose: Fluoroquinones(FQs),as one of the “Three Major” antibiotics widely used worldwide,have good pharmacokinetic characteristics and therapeutic effects in the treatment of bacterial infections in humans and animals.However,bacterial resistance and adverse reactions have also occurred one after another,which has adverse effects on ecosystem,human and animals' health.Chemiluminescence(CL)has been widely applied to the detection of FQs because of its high sensitivity,low background interference and simple operation.However,due to the fact that the study of its luminescence mechanism is not yet clear,or the detection of the target compounds of FQs is relatively single,the luminescence mechanism is partial and the study of homologues is lacking.Therefore,further research and discussion are needed.Methods: In this study,we mainly studied the chemiluminescence of eight fluoroquinolones in the peroxynitrite system(ONOOH),starting from the difference in the chemiluminescence spectra of FQs,using electron spin resonance and liquid chromatography-Mass spectrometry was used to investigate the mechanism of chemiluminescence,to further explore the excited state intermediates in the luminescence reaction,and to elucidate the mechanism of the chemiluminescent diversity of ONOOH and FQs,providing new ideas for the mechanism study of the chemiluminescence reaction system.Results: The results of the study demonstrate many new features of fluoroquinolones in chemiluminescence spectroscopy,chemiluminescence quantum yield,and reaction schemes.When comparing the distribution of chemiluminescence spectra and fluorescence spectra of FQs,the diversity of the differences between the drugs can also be observed.The results show that the difference between different fluoroquinolones may be due to the difference in the 8-position substituents;at the same time,a free-radical capture experiment was conducted to clarify the existence of free radicals during the reaction and to clarify that the free radicals are hydroxy free Base,suggesting that the presence of hydroxyl radicals and excited peroxynitrite may explain these observations;moreover,by studying the defluorination rate and the relationship between the defluoridation rate and the intensity of chemiluminescence,the substance itself is indirectly reflected.The chemical structure and its fluorescence quantum yield will have a greater impact on chemiluminescence.Finally,the stable products after the reaction were isolated and identified,further demonstrating the chemiluminescence reaction mechanism of fluoroquinolones.Conclusion: The diversity of fluoroquinolone chemiluminescence is closely related to the two factors of hydroxyl radical and excited peroxynitrite.This discovery helps to change people's inherent thinking of the chemiluminescence phenomenon,and to further understand the mechanism of chemiluminescence is of great significance.
