Objective: To observe the chinese medicine Shengjiangsan on diabetic rat intestinal bacteria(Escherichia coli,Streptococcus,intestinal Lactobacillus and Bifidobacterium)the impact of structural changes and related inflammatory factors,to explore the correlation of shengqingjiangzhuo theory and the pathogenesis of diabetes mellitus,and provide experimental basis for clinical application.Methods:1.Take 70 SPF male SD rats,weight 140-160 g,after 1 weeks of feeding,according to 1:6 were divided into normal control group and model group,model group fed with high fat diet for 8 weeks after intraperitoneal injection of small dose of streptozotocin(STZ)diabetic rat model induced by 30mg/Kg,excluding minors model and death of10 rats,then the rats 50 were randomly divided into model group,acarbose group,low dose group,Shengjiangsan middle dose group,high dose group,were divided into 6groups,10 rats in each group.2.Before the treatment,model group and acarbose group and Shengjiangsan low,middle and high dose group compared with normal control group,FBG,IL-6,TNF-,FINS,intestinal alpha streptococci and Escherichia coli were significantly increased(P<0.01),the number of bifidobacteria and lactobacilli were significantly decreased(P<0.01)before treatment;after the normal control group and model control group each index had no significant difference(P>0.05).3.according to the corresponding dose of drug intervention for 6 weeks,the rats weight 2 times weekly said,according to the changes of body weight adjusted dose,and observe the general condition of rats,second,4 and 6 weeks of rat tail vein blood to measure fasting blood glucose(FBG);sixth weeks the rats were sacrificed,the rats were taken from each group only 8 samples by enzyme-linked immunosorbent assay(ELISA)detection of interleukin-6(IL-6),tumor necrosis factor alpha(TNF-alpha),fasting insulin(FINS)and other indicators,real-time fluorescence quantitative(Realtime PCR)bacterial quantitative determination technology,statistics and data analysis.Results:1.the general condition of the rats: the rats in the normal group were in good mental state,the reaction was sensitive,the diet and urine volume were normal,the hair color was bright,and the weight was increased steadily.Other groups were significantly thinner after STZ injection,and the emergence of polyphagia,polyuria,listlessness,hair color is yellow and dull and other symptoms,part of the tail vein of the rats appeared obvious purpura.2.before the treatment,model group and acarbose group and Shengjiangsan low,middle and high dose group compared with normal control group,FBG,IL-6,TNF-,FINS,intestinal alpha streptococci and Escherichia coli were significantly increased(P<0.01),the number of bifidobacteria and lactobacilli were significantly decreased(P<0.01)before treatment;after the normal control group and model control group each index had no significant difference(P>0.05).3.acarbose group and Shengjiangsan low,middle and high dose group in second weeks,FBG decreased,but compared with the model control group and before treatment was not statistically significant(P>0.05);gradually decreased after 4 weeks and 6 weeks of FBG were lower than at the same time,the model control group,the difference was statistically significant(P<0.01).Shengjiangsan in high dose group and acarbose group at 4 weeks and 6 weeks FBG than before treatment decreased,with statistical significance(P<0.05),while the Shengjiangsan low dose group than before treatment has decreased,but there was no statistical significance(P>0.05).The model control group with the course of growth,blood glucose increased,but no significant difference(P>0.05),acarbose Shengjiangsan group,low dose group,middle dose group and Shengjiangsan Shengjiangsan and there were no significant differences between the groups of high dose group at different time points(P>0.05).4.after treatment,acarbose group and Shengjiangsan low,medium and high dose group of IL-6,TNF-alpha and FINS levels before treatment and model group were significantly decreased(P<0.01),but at the same time were higher than the normal control group(P<0.01);Shengjiangsan low,medium and high dose group IL-6 TNF-,FINS,alpha level compared with acarbose group were lower(P<0.05),especially in lifting powder IL-6,TNF-dose group alpha and FINS were significantly lower(P<0.05).5.after treatment,acarbose group and Shengjiangsan low,medium and high dosegroup of Escherichia coli than before the treatment and model group were significantly reduced(P<0.05),but still higher than the normal control group(P<0.05),acarbose group and Shengjiangsan low,medium and high dose group group no significant difference(P>0.05);compared with the normal control group,Shengjiangsan number of low and high dose group enterostreptococcus increased,but no statistical difference(P>0.05),acarbose group and Shengjiangsan dose group decreased,with statistical significance(P<0.05),at the same time the treatment group the number of intestinal Streptococcus compared with model control group and before treatment were significantly reduced(P<0.01).6.after treatment,compared with the normal control group,acarbose group and Shengjiangsan low,medium and high dose group significantly increased the number of bifidobacteria(P<0.01),while Lactobacillus in Shengjiangsan dose group and acarbose group increased significantly(P<0.05);acarbose group and lifting scattered low,medium and high dose group of Bifidobacterium and Lactobacillus number compared with model control group and before treatment were significantly increased(P<0.01).Conclusion:1.Shengjiangsan can improve the general state of DM rats,reduce serum INS,blood glucose levels,indicating that the increase in insulin secretion and insulin resistance;2.The treatment of DM can reduce the levels of IL-6 and TNF-alpha in rats with DM,and its effect is better than that of acarbose,which indicates that Shengjiangsan can significantly improve the inflammatory state of rats;3.Shengjiangsan can significantly increase the number of Lactobacillus and bifidobacteria in DM rats,reduce the number of harmful bacteria Escherichia coli and Streptococcus,adjust the intestinal micro ecological environment.
