Objective To investigate the effect of silencing Nanog gene on the proliferation,migration and invasion of esophageal squamous cell carcinoma TE-1 cells and to explore the regulatory mechanism.Methods The green fluorescent marker specific Nanog-si RNA was transfected into TE-1 cells by liposome Lipofectamine2000,and the expression of fluorescent protein was observed by fluorescence microscopy,and the transfection results were detected by real-time,PCR and Western blotting;CCK-8 method was used to detect the proliferation of TE-1 cells;influence on the migration and invasion of TE-1 cells by silencing Nanog detection of Wound Healing and Transwell experiment after the gene expression of m RNA and protein;real-time PCR and Western-blotting detection after silencing the Nanog gene.Results Experiments showed that Nanog silencing can reduce the inhibition of TE-1 Esophageal Squamous Cell and induces cell migration and invasion(P< 0.05);The levels of m RNA and protein in Stat3,p-Stat3,VEGF and ?-catenin decreased(P< 0.05).ConclusionsSilencing Nanog can effectively inhibit the proliferation,migration and invasion of esophageal cancer TE-1 cells,and the mechanism is that the silencing of Nanog gene is regulated by the Stat3 pathway.
