ObjectiveThis study was to investigate the protect effect and mechanism of sinomenine on myocardial ischemia reperfusion injury.MethodsMyocardial ischemia reperfusion injury model in rats was established by coronary artery ligation.50 SD rats were divided into 5 groups according to the principle of randomization as follows:(1)sham operated group,(2)model group,(3)15 mg/kg of Sinomenine group,(4)30 mg/kg of Sinomenine group,(5)60 mg/kg of Sinomenine group.After thoracotomy,only the sham operation group unligated,the other group of rat ligated left coronary artery 30 mins and then reperfused 120 mins.The ischemia reperfusion model had been established.Blood and heart tissue were taken immediately to measure the following indicators: The area of myocardial infarction was measured by nitro blue tetrazolium chloride(NBT)staining assay;Myocardial apoptosis was measured by TUNEL method;The content of TNF-?,IL-1?,CK-MB and c Tn I were determined by ELISA;The expression of NF-?Bp65 and TLR4 was determined by Western blot.ResultsCompared with sham operation group,the infarct size,cardiomyocytes apoptosis and the content of TNF-?,IL-1?,CK-MB and c Tn I were increased,and TLR4/p65 signal was up-regulated in model group.In rats,which are pretreated with Sinomenine(30 mg/kg,60 mg/kg),the injury of myocardial cells induced by ischemia-reperfusion can be obviously improved.The content of TNF-?,IL-1?,CK-MB and c Tn I were decreased,and inhibited the up-regulated of the TLR4/NF-?B p65 signal.Compared with the model group,the myocardial infarction area of were decreased in sinomenine group,the differences were statistically significant(P<0.01).ConclusionsSinomenine pretreatment can inhibite the up-regulation of TLR4/NF-?B p65 signaling pathway,which may be related to inhibit the release of inflammatory factor.The ischemia reperfusion injured myocardial cell can be protected by this way.
