ObjectiveTo evaluate the effect of quercetin on the activity of adriamycin against breast cancer and to explore its related mechanism.Methods1.The effects of quercetin,adriamycin,adriamycin combined with quercetin on the proliferation of human breast cancer cells MCF-7 and MCF-7/dox were detected by MTT assay.2.Flow cytometry was used to detect the apoptosis of MCF-7 and MCF-7/dox cells induced by adriamycin before and after quercetin intervention.3.Transwell was used to detect the invasion inhibition before and after quercetin intervention adriamycin on MCF-7 and MCF-7/dox cell.4.Flow cytometry detect cytotoxicity of adriamycin on BCSCs in MCF-7and MCF-7/dox cells before and after quercetin intervention.5.Flow cytometry was used to detect the changes of adriamycin accumulation in MCF-7 and MCF-7/dox cells before and after quercetin intervention.6.Western Blot method was used to detect the expression of ABC transporters p-gp,BCRP and MRP1 in MCF-7 and MCF-7/dox cells before and after adriamycin combined with quercetinResults1.Quercetin dose-dependently inhibited the proliferation of MCF-7 and MCF-7/dox;0.7?M quercetin had no effect on the above cell growth.2.Quercetin obviously increased the inhibitory effect of adriamycin on the proliferation of MCF-7 and MCF-7/dox cells,and sensitization fold increased by1.38 in MCF-7 cells and in MCF-7/dox cells by 1.73,compared with the single adriamycin group had evident statistical significance(P<0.05).3.Quercetin significantly enhanced the apoptosis induced by adriamycin on MCF-7 and MCF-7/dox cells(P<0.05).4.Quercetin significantly increased the invasion inhibition of adriamycin on MCF-7 and MCF-7/dox cells(P<0.05).5.Quercetin significantly enhanced the cytotoxicity of adriamycin on BCSCs(P<0.05).6.Quercetin significantly increased the amount of adriamycin accumulation in MCF-7 and MCF-7/dox cells(P<0.05).7.adriamycin combined with quercetin significantly down-regulated the expression of efflux ABC transporter include p-gp,BCRP and MRP1 on MCF-7and MCF-7/dox cells compared with the adriamycin group(P<0.05).ConclusionQuercetin can significantly enhance the anti-breast cancer activity of adriamycin,and its mechanism is related to cooperate with adriamycin to kill BCSCs and down-regulated the expression of efflux ABC transporters include p-gp,BCRP,MRP1.
