BackgroundBecause of the emergence of drug resistance in tumor cells,chemotherapy limits the therapeutic effect and prognosis of the patients with ovarian cancer.Taxol is a kind of widely used chemical drug in tumor treatment,but the primary or secondary drug-resistance caused by Taxol is the main factor that influences the effect of chemotherapy,which is also the main cause of tumor recurrence,and the molecular mechanism remains unclear.Previous studies have shown that the abnormal expression of long non-coding RNAs(lncRNAs)can contribute to the occurrence and prognosis of cancer,and whether lncRNAs are related to paclitaxel-resistance in ovarian cancer remains to be studied.ObjectiveThe aim of this study is to establish the paclitaxel-resistance ovarian cancer cell model,further to analyze the relation between the three lncRNAs and paclitaxel-resistance of ovarian cancer cells.Methods1.The paclitaxel-resistance cells A2780/Taxol were established using the different paclitaxel concentration gradient.2.The expression of three selected kinds of lncRNAs,including n375709,n377806,n369241,were detected by real-time fluorescent quantitative reverse transcription PCR technology(qRT-PCR)technology.3.The expression of n369241 lncRNAs was knocked down by RNAi technology and the sensitivity changes of paclitaxel resistance in ovarian cancer cells were observed.Results1.The paclitaxel-resistance cells Skov-3 /Taxol and A2780/Taxol were successful established by using the concentration gradient.The semi inhibitory concentration of IC50 was 6.64 + 0.069 g/ml,and the multidrug resistance index was 25.54.In addition,a strong cross resistance was also produced for adriamycin(ADR)and 5-fluorouracil(5-FU).2.qRT-PCR results showed that the expression of n375709,n377806 and n369241 were down-regulated in A2780/Taxol cell compared with A2780 cell(P<0.05),and the highest down-regulated lncRNA was n375709(4.35-fold).n377806 and n369241 were down-regulated by 2.0 times and 1.85 times(P < 0.05).3.Down-regulatedn of 369241 lncRNAs expression increased paclitaxel-resistance in ovarian cancer cell.Conclusion1.Abnormal expression of three kinds of lncRNAs was associated with paclitaxel-resistance in ovarian cancer cell.2.The n375709 lncRNAs expression differences is related with paclitaxel-resistance in ovarian cancer cell.
