Keloid is a fibroblast hyperplastic disease caused by abnormal proliferation of fibroblasts and excessive deposition of extracellular matrix collagen in the process of skin wound repair[1].The clinical manifestations of keloid are more than the original lesions and higher than the benign masses of normal skin.Some patients may be accompanied by local pruritus pain,and can also affect local physiological function when contracture occurs.Keloid is a benign mass,but because of its serious impact on beauty,it has a negative effect on the patient's physiology and psychology.The quality of life of the patients was greatly reduced[2].However,there is no significant way to improve symptoms and reduce recurrence in western medicine treatment at present.Chinese gallnut scar Ointment(Black vinegar,No.5)A series of studies were carried out on the treatment of cicatrix with Chinese medicinal ointment(black vinegar,gallnut,Scolopendra,honey and plum blossom).However,the mechanism of its inhibition on fibroblast proliferation is not clear.Based on previous studies,we further studied the mechanism of gallnut scar ointment inhibiting the proliferation of keloid fibroblasts,so we can offer a theoretical basis for the treatment of keloids.ObjectiveTo detect the expression of miR-21 in normal human skin and keloid tissues,and to intervene the keloid tissue of nude mice with gallnut scar ointment.To observe the effect of gallnut scar ointment on the expression of miR-21/mTOR signal pathway and miR-203 in keloid tissues of nude mice,and to explore the mechanism of gallnut scar ointment in treating keloid from the point of view of miR-21/mTOR signaling pathway miR203.So as to provide more theoretical support for the clinical application of Galla chinensis scar ointment for keloid treatment.MethodsIn this study,18 nude mice were transplanted with keloid tissue after operation to establish keloid model,and were randomly divided into experimental group and control group after 14 days of drug intervention,the experimental group was smeared with Galla chinensis scar ointment.The expression levels of mir-21 and mir-203 in keloid tissues were detected by RT-PCR technique 28 days later,and the expression levels of mir-21 and mTOR in keloid tissues after intervention were detected by immunohistochemical method.The work of the immunohistochemistry section is handled by the same door.This paper only describes the results,the specific experimental process and data can be found in the same paper.ResultsIn The expression level of miR-21 in the experimental group was significantly lower than that in the control group(P<0.05)and the expression level of PTEN in the control group was significantly higher than that in the control group(P<0.05).The levels of miR-21 and mTOR in the experimental group were significantly lower than those in the control group(P<0.05).ConclusionChinese gall scar ointment can be used to treat keloid effectively and has good clinical effect.It is found that the therapeutic effect of Galla chinensis scar ointment on keloid may be achieved by regulating miR-21/mTOR signaling pathway and miR-203 expression level.
