Objective1.To investigate the safety and efficacy of the gastric subserous as a transplanted islets site2.To compare the transplanted efficacy of gastric subserous islets transplantation and portal vein islets transplantation in the treatment of diabetic rats;3.Find the most suitable site for islet transplantation.MethodsSTZ was used to induce type 1 diabetes mellitus in rats with islet transplantation.The diabetic rats were randomly divided into three groups: the gastric subserous group,the portal vein group,the control group.Type XI collagenase was injected through choledoch to digest the pancreas,and the islets were isolated and purified by islets.The specificity of the islet was judged by DTZ and counted IEQ and islets purity.The activity of islets was determined by trypan blue test.500 IEQ fresh islets were transplanted into the gastric subserous and into the portal vein,respectively.The control group was infused with the same amount of RPMI1640.After islet graft in the diabetic rats,the changes in blood glucose,intraperitoneal glucose tolerance and assessment the function of islets.ResultThe number of islets was(310 ± 42),and the purity of islets(89 ± 4.52)%was obtained by DTZ staining and the activity of islets was(89.08 ± 3.76)% was obtained by Trypan blue staining.The glucose level of in the gastric subserous and portal vein diabetic rats was significantly decreased compared with that before transplantation(P<0.05;P<0.05).The blood glucose was a significant difference by the control group(P<0.05;P<0.05).There was no significant difference in the glucose between the gastric subserous and portal vein after blood glucose transplantation(P<0.05).The effective control of blood glucose in gastric subserous and portal vein was(18±1.87、11.6±2.5)days(P<0.05).The gastric subserous and portal vein rats had good intraperitoneal glucose tolerance(P<0.05).ConclusionGastric subserous as a islet transplantation site is safe and effective;Short-term,the gastric subserous islet transplantation treatment of diabetic rats effect was higher than that of portal vein.Compared with portal vein islet transplantation,the operation of pancreatic islet transplantation is more simple and safe,and it can be considered as one of the most suitable transplant sites for islet transplantation.
