ObjectiveTo investigate the effect of Rutin(RT)combined with oxaliplatin(OXA)on the proliferation,cell cycle and apoptosis of human gastric cancer cell SGC-7901 and the possible role of p38 MAPK signal transduction pathway.MethodsThe human gastric cancer cell line SGC-7901 in logarithmic growth phase was taken as the research object.The effect of p38 inhibitor(SB203580),rutin and oxaliplatin on the proliferation of human gastric cancer cell SGC-7901 was detected by MTT assay.Hoechst33258 nuclear staini ng was used to observe the morphological changes.Cell cycle and cell apoptosis were detected by flow cytometry.The expression of p38,p-p38,Bcl-2,Bax,pro-caspase3,cleaved-caspase3,cleaved-caspase7,cleaved-ca spase8 and cleaved-caspase9 protein expression in SGC-7901 cells were analyzed by Western-blot.ResultsP38 inhibitor could promote SGC-7901 cells proliferation(P<0.05).Either rutin or oxaliplatin could inhibit the proliferation of SGC-7901 cells,and the inhibitory effect of their combination was more significant.;p38 inhibitors promoted cell proliferation by accelerating the G0/G1 phase,whereas rutin,oxaliplatin and combination groups the cells were arrested at G0/G1 and induced apoptosis;the expression of p-p38,cleaved-caspase3,cleaved-caspase7,cleaved-caspase8,cleaved-caspase9 and Bax in the p38 inhibitor group was significantly lower than that in the control group.The expression of Bcl-2 was up-regulated,which resulted in a significant increase in Bcl-2/Bax levels(P < 0.05).After the action of rutin and oxaliplatin,p-p38,cleaved-caspase3,cleaved-caspase7,and claved-The expression of caspase8,cleaved-caspase9 and Bax protein was significantly increased compared with the blank control group,and the expression of Bcl-2 was down-regulated,and the Bcl-2/Bax level was significantly lower(P < 0.05).Rutin and oxaliplatin After the combined action,the above protein change trend alone with rutin,oxaliplatin The same but more significant effect(P <0.05).ConclusionsBoth rutin and oxaliplatin could inhibit the proliferation and induce the apoptosis of SGC-7901 cells,which may be related to the activation of p38 MAPK pathway,the down-regulation of Bcl-2 / Bax and the up-regulation of the expression of caspase-related proteins.The joint application of a certain synergy.
