| Objective:To study the expression of DAB2IP in colorectal cancer and its effect on colorectal cancer EMT;to analyze the correlation among DAB2IP,ZEB1,and E-cadherin in colorectal cancer;to investigate whether there is some signaling mechanism between DAB2IP and ZEB1 to regulate tumor EMT;and to provide a new idea for the molecular mechanism of DAB2IP involved in the invasion and metastasis of colorectal cancer.Methods:The expression,clinical significance and prognostic analysis of DAB2IP in colorectal cancer tissues were obtained through Oncomine database.By interfering with the expression of DAB2IP,real-time quantitative PCR and Western blotting were used to detect whether the expression of EMT-related markers changed,and the effect of DAB2IP on EMT in colorectal cancer was studied.The expression of DAB2IP,ZEB1,and E-cadherin protein was detected by immunohistochemical staining.The presence of differential expression in the normal colorectal tissues and colorectal cancer tissues was observed,further understand the relationship between the three and colorectal cancer clinical pathological parameters,and analyze the correlation among the three.Results:1.The expression of DAB2IP mRNA in colorectal cancer tissue was lower than that in normal colorectal tissue,it was decreased with the increase of histological grade,and the low expression of DAB2IP may predict the poor prognosis of patients,suggesting that DAB2IP plays an inhibitory role in the development of colorectal cancer.2.After the interference of DAB2IP,the expression of epithelial phenotype marker E-cadherin decreased,the expression of interstitial phenotype marker Vimentin increased,the expression of EMT transcription factors ZEB1,Snail and Slug increased,suggesting that interference with DAB2IP could promote colorectal cancer EMT.3.The expression of DAB21P protein in colorectal cancer was lower than that in normal colorectal tissue,and the expression of DAB2IP protein decreased with the increase of histological grade and clinical stage,suggesting that the down-regulation of DAB2IP expression plays an important role in the occurrence and development of colorectal cancer.4.The expression of ZEB1 protein in colorectal cancer was higher than that in normal colorectal tissue,and the expression of ZEB1 protein increased with histological grade and clinical stage,suggesting that the up-regulation of ZEB1 expression plays an important role in the occurrence and development of colorectal cancer.5.The expression of E-cadherin protein in colorectal cancer was lower than that in normal colorectal tissue,and the expression of E-cadherin protein decreased with the increase of histological grade and clinical stage,suggesting that the down-regulation of E-cadherin expression plays an important role in the occurrence and development of colorectal cancer.6.The expression of DAB2IP in colorectal cancer tissue was negatively correlated with ZEB1,the expression of DAB2IP was positively correlated with E-cadherin,and the expression of ZEB1 was negatively correlated with E-cadherin,suggesting that there may be some signaling mechanism between DAB2IP and ZEB1 that causes EMT in tumor cells.Conclusion:Interference with DAB2IP could promote colorectal cancer EMT.DAB2IP,ZEB and E-cadherin are important for the occurrence and development of colorectal cancer,and the expression of the three is correlated in colorectal cancer. |
