Objective: Acne vulgaris is a common chronic inflammatory follicular sebaceous gland disease.This disease,especially lesions of severe acne vulgaris such as cyst and nodule,brings serious face damage to patients,which has huge impact on patients' psychology and social interaction.At present,many scholars think that acne vulgaris is a multi gene disease which is interacted by genetic and environmental factors.Therefore,this study intended to investigate the gene polymorphism of TIMP-2 and IL-1? in patients with acne vulgaris in Han Chinese of Sichuan.Methods : A total of 394 individuals were enrolled,including 104 healthy controls and 290 patients with acne vulgaris(98 mild,109 moderate and 83 severe).1.To investigate each individual,an unified designed acne vulgaris questionnaire was adopted and filled in by each individual.2.After extracting the peripheral venous blood 5mL from each individual,extracting the leukocyte DNA and obtaining the target gene,we detected the SNP of the TIMP-2 gene at position rs8179090 and the IL-1? gene at position rs1800587 of the acne vulgaris patients and healthy controls by the method of gene sequencing and SNaPshot typing.Results: 1.The morbidity rate of acne vulgaris in first-degree relatives with moderate and severe acne vulgaris patients was significantly higher than that in the healthy group in Han Chinese of Sichuan(P < 0.05).The highest morbidity rate of first-degree relatives was in patients with severe acne vulgaris.2.The awareness rate of the acne vulgaris as a disease in moderate and severe acne vulgaris patients was significantly higher than that in the healthy and mild group(P < 0.05).3.The proportion of patients with moderate and severe acne vulgaris considering that acne vulgaris had a great impact on work and life was significantly higher than that of mild acne vulgaris patients(P < 0.05).The highest proportion was in patients with severe acne vulgaris.4.The independent risk factors associated with acne vulgaris were: bad mood(OR value 8.466),first-degree relatives with acne vulgaris(OR value 2.897),fatty food(OR value 2.513),high glycemic food(OR value 2.432),washing face without facial cleanser(OR value 2.365),spicy food(OR value 2.332).5.The SNP of TIMP-2 gene at position rs8179090 in acne vulgaris patients and healthy controls were significantly different(P < 0.05).The presence of C allele would increase the risk of acne vulgaris,but there were no significant differences between mild,moderate and severe acne vulgaris patients(P > 0.05).6.The SNP of IL-1? gene at position rs1800587 in acne vulgaris patients and healthy controls showed no significant difference(P > 0.05)and there were no significant differences between mild,moderate and severe acne vulgaris patients too(P > 0.05).Conclusion: 1.The morbidity rate of acne vulgaris in first-degree relatives with moderate and severe acne vulgaris patients was significantly higher than that in the healthy group in Han Chinese of Sichuan.Patients with severer acne had a higher morbidity rate in first-degree relatives.2.Patients with moderate and severe acne vulgaris had a better awareness rate that acne was a disease than healthy and mild acne vulgaris patients.3.The people with severer acne thought the disease would make greater influence on work and life.4.Bad mood,family history,unhealthy diet and improper nursing methods had an impact on the morbidity rate of acne vulgaris.5.The SNP at the rs8179090 position of the TIMP-2 gene was associated with acne vulgaris,but it was uncorrelated with the severity of acne vulgaris;the presence of the C allele at the rs8179090 position of the TIMP-2 gene would increase the risk of acne vulgaris.6.The SNP at the rs1800587 position of the IL-1? gene was not associated with acne vulgaris and was not related to the severity of acne vulgaris.
