| In recent years,the frequent occurrence of allergic diseases has caused serious problems in the normal life of allergic people,and the attention of governments and researchers around the world to allergic diseases is increasing.For controlling the growth of allergic diseases,developing new drugs and functional food has become a hot topic in recent years.Studies have shown that marine algae polysaccharides and deep-sea bacteria metabolites have various biological activities.Exploring the effects of algae polysaccharide and deep-sea bacteria metabolites on allergic reactions can provide new ideas for the prevention and control of allergic diseases,and also provide a theoretical basis for the development of anti-allergic food or drugs.In this study,PHPS(Hp+Vc)was obtained from the polysaccharides(PHPS)of Porphyra with high pressure and ascorbic acid method.The molecular weight decreased from 2,960,240 Da to 132,958 Da,the viscosity decreased from 1.76 dL/g to 0.14 dL/g,and the ratio of reducing sugar increased by 23.59%.The infrared spectra showed that the basic chemical structure of PHPS(Hp+Vc)did not change,and it still contained the characteristic absorption peak of the active group sulfate,and the content of sulfate was 11.41%.In this study,anti-allergic activities of PHPS and PHPS(Hp+Vc)were evaluated by the established cell model in vitro and animal model in vivo,and the anti-allergic mechanism of PHPS and PHPS(Hp+Vc)was explored.In vitro cell model analysis showed that PHPS and PHPS(Hp+Vc)could inhibit the degranulation and histamine release of rat basophil(RBL-2H3)and bone marrow derived mast cells(BMMCs),and PHPS(Hp+Vc)inhibition activity was higher than that of PHPS.The results of flow cytometry showed that PHPS and PHPS(Hp+Vc)have no influenc of mast cells apoptosis.In vivo animal model analysis showed that PHPS and PHPS(Hp+Vc)can relieve the allergic symptoms such as low temperature and diarrhea in mice,and reduce the level of specific IgE,histamine and mast cell protease in the serum of allergic mice,reduce the level of Th2 related cytokines in serum,increase the level of Treg related cytokines and the level of interleukins-21;Cytometry results showed that PHPS and PHPS(Hp+Vc)could up-regulate the proportion of regulatory T(Treg)cells in splenic lymphocytes of allergic mice.The results showed that PHPS and PHPS(Hp+Vc)could alleviate allergic reaction by governing the level of allergen related cytokines and the proportion of Treg cells.In this study,24 compounds with very small molecular weight were isolated from the Williamsia sp.MCCC 1A11233 metabolites of the deep sea actinomycetes,named CDMW-1~CDMW-24.Anti-allergic activity was evaluated by cell model in vitro and mouse model in vivo.CDMW-2,CDMW-3,CDMW-5,CDMW-6,CDMW-8,CDMW-15 and CDMW-21 can inhibit the production of RBL-2H3 cells and BMMCs degranulation,histamine release and cytokine production.CDMW-3,CDMW-5 and CDMW-15 can induce the apoptosis of mast cells by inducing the inhibition effect in the cell model in vitro and passive skin allergy model in vivo.In conclusion,PHPS,PHPS(Hp+Vc)and CDMW-3,CDMW-5,CDMW-15 have anti-allergic activity,but the two mechanisms are different,which can provide theoretical support for the development of new anti-allergic food and drugs. |
