Effects Of Ira Maude On Regulatory B Cells And Cytokines

Objective:To study the effects of Ira Maude on collagen induced arthritis(CIA)and the changes of interleukin-10(IL-10),transforming growth factor(TGF-beta),Foxp3 and Bcl-6.Methods:30 healthy and clean female Wistar rats of 6-8 weeks old,body mass(180-220)g,were randomly divided into blank control group,model control group and Ira Maude group,and 10 rats in each group were tested.Both model control group and Ira Maude group were given CIA model of bovine type II collagen(C II)and complete Freund adjuvant(CFA)0.3ml/ only.The body weight changes of CIA rats were observed after 28 days of treatment.The pathological changes of synovial tissues were observed by HE staining,and the content of IL-10 and TGF-beta in serum was detected by ELISA method;real time fluorescence determination was used.PCR(RT-PCR)was used to detect the expression of IL-10 mRNA,TGF-beta mRNA and Foxp3 mRNA and Bcl-6mRNA in synovial tissues.Result:(1)after treatment,the body weight of the Ella mod group increased significantly and the temperament was temperate.The rats in the model control group had slow weight increase and irritability.(2)after the joint synovial membrane was stained with HE,the joint synovial cell layer in the Ira Maude group was 4 layers(normal at 1-3 layers),and no obvious hyperplasia and inflammatory cell infiltration were found.Model control The synovial cell layer increased to 10 layers,showing obvious proliferation and a large number of inflammatory cell infiltration.There was no significant difference in serum IL-10 content between Ella mod group and model control group(P >0.05).There was no significant difference in serum TGF-and P levels between Ella mod group and model control group(P>0.05).The level of IL-10 mRNA expression in the synovial membrane of the joint: there was no significant difference between the Ella mod group and the model control group(P>0.05),and the expression level of TGF-beta m RNA in the synovial synovium: the difference between the Ella mod group and the model control group was not statistically significant(P>0.05);the level of the expression of the spleen in the spleen was: the Ella mod group and the model Compared with the controlgroup,the difference was not statistically significant(P>0.05),and the expression level of Bcl-6mRNA in the spleen: there was no significant difference between the Ella mod group and the model control group(P>0.05).Conclusion:(1)after the treatment of Ira Maude,the weight of rats showed a significant trend of growth and temperate.(2)after the treatment of Ella mod,the expression of IL-10 and TGF-beta in the sera and joint synovium and the expression of Foxp3 and Bcl-6 in the spleen did not change significantly.(3)whether Ira Maude can influence the development of rheumatoid arthritis through other cytokines or signaling pathways after treatment with Breg remains to be further verified.
