Objective: To investigate the effects of bone marrow mesenchymal stem cells combined with amniotic membrane and magnesium alloy stent in the repair of osteochondral injury in non-bearing area of rabbit knee.Methods: Rabbit bone marrow-derived mesenchymal stem cells were isolated,cultured,passaged,and observed for growth.The flow cytometry cells were used to observe the expression of cell surface antigen to detecte the cells.The cells,amnion and magnesium alloy were co-cultured,and an experimental model of cartilage defect was made.The experiment was divided into three groups: group A was BMSCs + amnion + magnesium alloy group,group B was BMSCs + amniotic membrane group,group C was blank control group.Group A is the experimental group 1,group B is the experimental group 2,and group C is the control group.Three groups of implants were placed in animal models of cartilage injury.8 weeks and 12 weeks later,ross observation,micro-CT observation and histological observation were performed on each group.At the same time,weve the movement of animals in each group,cartilage injury repair situation,chondrocytes and magnesium alloy absorption situation.At the same time,the movement of animals,cartilage injury repair situation,chondrocytes and magnesium alloy absorption situation in each group.Results: 1.The extracted bone marrow cells grow logarithmically after 1-2 days of incubation.The expression of CD34(-),CD45(-),CD44(+),CD90(+)and CD106(+)in bone marrow cells were detected by flow cytometry,so the extracted cells proved rabbit BMSCs.2.Magnesium alloy and bone marrow mesenchymal stem cells co-culture,and made bone marrow mesenchymal stem cells attached to the magnesium alloy.3.At the 8th week and the 12 th week respectively,three groups of experimental rabbits were tested.A group of cartilage injury was observed in a large number of cartilage tissue filling group B cartilage injury was part of cartilage tissue filling group C was a large number of fibrous tissue filled.Histological observation showed that cartilage injury departments were densely chondrocytes in group A at 8 and 12 weeks,and the number of chondrocytes at 12 weeks was more than 8 weeks.Magnesium alloy is partially absorbed in group A at 8 and 12 weeks,12 weeks to more than 8 weeks.In group B,a small amount of chondrocytes were formed in the cartilage lesions at 8 and 12 weeks,similarly,the number of chondrocytes at 12 weeks was more than 8 weeks.Cartilage injury are a large number of fibrous tissue formation in group C at 8 and 12 weeks.Conclu Sron: Bone marrow mesenchymal stem cells combined with amniotic membrane and magnesium alloy stents can promote the generation of cartilage cells and contribute to the repair of cartilage damage.
