Objective:Using isotope labeled relative and absolute quantification(iTRAQ)tech nology screening of esophageal squamous cancer radiotherapy and chemotherapy eff icacy and survival of serum differential proteins,and guiding the individualized treat ment of clinical treatment.Methods:1.Included 52 patients with definite diagnosis of local and advanced esophageal squamous cell carcinoma,standardized radiotherapy,j udgment efficacy and long-term follow-up,and collected patients before and after tre atment.2.Using iTRAQ technology to screen and radiate the therapeutic effect and prognosis of relevant differential proteins.3.Using Uniprot,KEGG and other database s to conduct biological analysis of the differential protein,and then screened the do minant protein with relevant pathway mediated by tumor related,core protein.Result s:1.Follow-up results of short-term follow-up:59.62%(31/52)patients with esophageal cancer(CR+PR+SD)and 40.38%(21/52)patients with resistance to radiochemothera py(PD).Follow-up results of long-term follow-up:1?OS<2 years group 46.15%(24/52cases);2?OS<3 years group 32.69%(17/52cases);OS?3 years group 21.15%(11/52 cas es).2.Radiochemotherapy-sensitive group vs.radiochemotherapy-resistant group:A tota l of 17 significantly differentially expressed proteins were screened out,among whic h were up-regulated proteins ADIPOQ(adiponectin),Ig L C7,TF(serum transferrin),D EFA1(neutrophil defense A total of four factors,13 down-regulated protein PIGR(pol ymer immunoglobulin receptor),TF(serum ferritin),CD14(monocyte differentiation an tigen 14),etc.14.3.Differential protein screening among different survival periods:(1)A total of 42 significant differentially expressed proteins were identified,of which 19 were up-regulated and 23 were down-regulated;(2)Differential proteins that were present at the same time between the two groups had 13 One,which simultaneouslyupregulates 1 protein(SAA2 serum amyloid A-2 protein)and down-regulates 3 pro teins(SERPINA5 plasma serine protease inhibitor,HIST1H4 A histone H4,HIST1H3 A histone H3.1).By KEGG pathway analysis,TF,CD14,H4,H3.1 may be involved in t he regulation of HIF-1 signaling pathway,MAPK signaling pathway,Viral carcinogen esis,and Transcriptional misregulation in cancer signal pathway,respectively.Conclusio ns:1.The potential predominant proteins predicting the efficacy and survival of esop hageal squamous cell carcinoma can be screened from the peripheral blood and inv olved in some key pathways;2.TF,ADIPOQ,CD14,HIST1H4 A,HIST1H3A,SERPINA5 for esophageal squamous cell carcinoma Chemotherapy may have potential efficac y and survival prediction value.
