| Objective:1.To observe the effect of PGE1 on general condition and wound healing rate in scalded rats,and evaluate the clinical effect of PGE1 on wound healing.2.To observe the variation of PGE1 on mRNA level and protein level of Wnt1,β-catenin in skin tissue of scalded rats,and explore the mechanism of promoting effect of PGE1 about wound healing.To provide theoretical foundation for clinical application of PGE1,and a new idea for the treatment of burn wound.Methods:48 SPF level of adult healthy SD rats(weight 200+20g),with a male-female ratio of 50—50,provided by laboratory animal science department of Nanchang University.Randomly divided these rats into 4 groups(Random digital table method): False burn group(blank group,n=12),burn control group(control group,n=12),LiCl group(n=12),PGE1 treatment group(PGE1 group,n=12).After adaptive feeding for one week,anesthetize all rats by given intraperitoneal injection of 4% chloral hydrate(7ml/kg),blank group treated with 37℃ warm water after dorsal hair removing,the other three groups were treated by steam experiment instrument,set as 0.03 Mpa,106 ℃,5S,leading to 30% TBSA deep II °scald in dorsal sides after dorsal hair removing.After injury,the rats were immediately given anti-shock treatment of intraperitoneal injection of Ringer lactate solution.PGE1 group was giving PGE1(2.5g/kg)through caudal vein and LiCl group was giving Li Cl(15 mg/ kg)through caudal vein for 14 days,meanwhile the saline was giving in other groups.using iodophor to treat all scalded rats’ wound,BID.Observe general wound appearances and wound healing rate of each group at 7,10,14 d.Extracted the total mRNA and total protein of skin tissue of rats at 14 d,Using RT-PCR and Western blot to detect mRNA level and protein level of Wnt1,β-catenin in skin tissue of rats.Results:1.The appearance of the scald wound:The wound of Blank group rats graduallyshowed hair growth.The wounds of scalded rats gradually formed dry eschar,and healed under-scab.In PGE1 group,compare to Li Cl group and Control group,the wound was drier and the color was more ruddy,and had less exudation,more hair growth and the wound healing was faster.Li Cl group also showed more hair growth and faster wound healing than control group.2.Wound healing rate: We observed,at the 7th day,there was no statistically significant difference about wound healing rate among Control group,Li Cl group and PGE1 group(P>0.05).Wound healing rate of PGE1 group were significantly higher than the LiCl group and control group(P<0.05)at 10 th and 14 th day.Compare to control group,Wound healing rate of LiCl group were significantly higher(P<0.05)at10th and 14 th day.3.The mRNA levels of Wnt1 and β-catenin: There were no difference between Li Cl group and PGE 1 group(P>0.05),Compare to Blank group and Control group,it showed a significantly increased in PGE1 group(p < 0.05).and Control group was significantly higher than Blank group(p<0.05).4.Wnt1,β-catenin protein levels: There were no difference between PGE 1 group and LiCl group(P>0.05),Compared to Blank group and Control group,PGE1 group showed a significant increase(p < 0.05).Control group was significantly higher than Blank group(p<0.05).Conclusions:1.After scalded,the mRNA levels and protein levels of Wnt1,β-catenin in the skin tissue of rats was higher than normal skin tissue.The activation of Wnt/β-catenin signaling pathway was found after tissue injury.2.PGE1 can accelerate wound healing,which its mechanism might be activating Wnt/β-catenin signal pathway by upregulating the expression of Wnt1 and β-catenin. |
