With the worldwide increasing incidence of lung cancer,lung cancer has become a malignant tumor with the highest mortality rate in the world and in China.The early symptoms of lung cancer are not quite obvious,most of the non small cell lung cancer patients have reached advanced stage at diagnosis,and the prognosis is poor.Due to the wide application of chemotherapeutic drugs in the treatment,the cells have formed a resistance to drugs.Development of high efficiency and low toxicity of targeted therapy drugs has become an important direction in lung cancer research.With the rapid development of nanotechnology and its application in the field of biology,nanomaterials have come into people’s sight as an efficient delivery vector.The development of new drugs with nanotechnology has become a new trend in drug research.MicroRNA(miRNA)is a series of endogenous noncoding RNA with a length of about 20-25 nucleotides.miRNA regulates gene expression by binding to the 3′ non-coding region of mRNA.A large number of studies have shown that the abnormal expression of miRNA is closely related to the occurrence,development and prognosis of lung cancer.MiRNAs have been considered as diagnostic markers of lung cancer and specific targets for the treatment of lung cancer.They play an important role in the early diagnosis,treatment,prognosis and pathogenesis of lung cancer.Treatment of lung cancer with the aim of regulating miRNA expression ushered in new opportunities.MiR-203,as a tumor suppressor niRNA,plays an important role in the occurrence and development of non-small cell lung cancer.Studies have shown that miR-203 inhibits cell proliferation by inhibiting the expression of target proteins,such as survivin and Bcl-2.MiRNA with strong antitumor activity can be loaded by nano-materials,which can not only reduce the degradation of miRNA molecules,but also achieve the targeted delivery to tumor tissues.It provides a new technique for the treatment of lung cancer.This paper takes PEI-PLA as the basic material,construct the nanoparticles encapsulated curcumin and miR-203,based on its morphology,entrapment efficiency,drug addition ratio and drug release characterization and optimization,screening and preparation has strong stability,ideal size,nano-drug and has good drug loading and release ability.The results of in vitro showed that the curcumin and miR-203 loaded with nano-materials could effectively escape the lysosome,and then quickly released into the cytoplasm of A549 cells.After the drug enters the cytoplasm,it can effectively down regulate the expression level of target genes survivin and Bcl-2,which can effectively inhibit the proliferation of cells and kill the tumor cells.This paper presents the design and encapsulation of curcumin and miR-203 nanoparticles were prepared,and study its biological effects on cell proliferation and the mechanism of action in A549 cells,providing a novel strategy for the treatment of non-small cell lung cancer.
