| Objective Patrinia heterophylla bunge.is a traditional Chinese medicine which could provide a cure for leukemia.Here,we mainly studied the effects P.heterophylla has on cell cycle.And further investigated the possible mechanism.Methods The HL-60 cells and K562 cells were treated for 24 and 48 hours with various P.heterophylla solutions which differed in concentration,to study the effects P.heterophylla has on cell cycle.Flow Cytometry and q RT-PCR ware employed to detect the changes of cell cycle status and expression of CDK1、Cyclin B1、CHK1、CDC25C m RNA respectively.Results P.heterophylla could arrest HL-60 cells and K562 cells in G2/M phase and we found that HL-60 and K562 cells in G2/M phase increased in a dose-dependent way when treated with P.heterophylla for 24 hours.For HL-60 cells,when treated with 10 mg/m L P.heterophylla for 24 hours,the proportion of G2/M phase showed much higher values than control(36.61% vs 10.27%,P<0.05).And for K562 cells,the proportion of G2/M phase with 20 mg/m L P.heterophylla was significantly higher than the control group(54.22% vs 14.7%,P<0.05).But it is worth to note that the proportion of cells in the G2/M phase after treated for 48 hours seemed no changes compared with 24 hours.(P>0.05).Besides these,The expression of cell cycle-correlated m RNA was also affected in K562 cells when treated with P.heterophylla,including the increased expression of CHK1 m RNA and the decreased expression of CDK1、Cyclin B1、CDC25C m RNA after treated with P.heterophylla both for 24 hours and 48 hours.Conclusion The P.heterophylla could block the cell cycle of HL-60 cells and K562 cells in G2/M phase.It could increase the expression of CHK1 m RNA and decrease the expression of CDK1、Cyclin B1、CDC25C m RNA in K562 cells. |
