Objective:By oxycodone pretreating to kidney ischemia-reperfusion model in rats,observed the renal pathological changes,renal function indexes,the expression of Bcl-2,the Bax and Caspase 3.To investigate the protective mechanism of hydroxyketone pretreatment for apoptosis during renal ischemia reperfusion in rats.Methods:Thirty-six male Wistar rats were randomly divided into three groups: Oxycodone preconditioning group(group Oxy),Ischemia-reperfusion group(group I/R),and Sham peration group(group S)(n=12 each).To establish the model of renal I/R injury,the left renal pedicles of group Oxy and group I/R were clamped with atraumatic clips for forty-five min,following by reperfusion,and the right rat kidney was removed immediately after onset of reperfusion.At fifteen min before reperfusion,oxycodone 0.5 mg/kg was injected into caudal vein in Oxy group,and the same amount of normal saline was injected in I/R group and Sham group,After twenty four hours reperfusion,the blood samples were drawed by cardiac puncture for concentrations measurement of blood urea nitrogen(BUN)and serum creatinine(Cr).The left renal were used for examination of pathological changes under light microscope.Using immunohistochemistry to determinate Bcl-2?Bax and Caspase-3 expression in renal tissues?Results:Compared with group S,group I/R and Oxy had obvious pathological damage.Compared with I/R group,Oxy group was less pathological.Compared with group S,concentrations of the serum creatinine and blood urea nitrogen in Oxy and I/R groups were significantly increased(P<0.05),and the Bcl-2/Bax of renal tissues was decreased and Caspase-3 of renal tissues was up-regulated at 24 h of reperfusion(P<0.05).Compared with the group I/R,concentration of the serum creatinine and blood urea nitrogen concentration were significantly decreased in the Oxy group after 24 h(P<0.05),the expression of the renal tissue Bcl-2 was up-regulated,Bax expression was down-regulated,Bcl-2/Bax increased,and Caspase-3 was down-regulated(P<0.05).Conclusion: Hydroxyketone pretreatment to relieve renal ischemia reperfusion pathology and renal function injury,and to regulate the balance of Bcl-2 and Bax,inhibit the expression of Caspase-3,thus to alleviate apoptosis.
